摘要
目的:研究温胆汤对精神分裂症模型大鼠行为学及其海马组织神经调节蛋白1(NRG1)、ErbB4 mRNA表达的影响。方法:将60只大鼠随机分为6组,即正常组、模型组、氯氮平20 mg/kg组、温胆汤40g/kg组、温胆汤20g/kg组、温胆汤10g/kg组。正常组和模型组分别ig生理盐水20 ml/kg,氯氮平20 mg/kg组ig氯氮平原药20 mg/kg,温胆汤40、20、10 g/kg组分别ig温胆汤生药40,20,10 g/kg,1次/d,共21 d。在末次给药2 h后,除正常组外,其余各组一次性ip MK-801 0.6 mg/kg,以诱发精神分裂症模型,造模后,观察并记录各组大鼠的行为学改变,观察3 d后,处死并取海马组织检测,观察大鼠的行为学改变,并做刻板行为和共济失调评分;RT-PCR检测大鼠海马组织NRG1、ErbB4 mRNA表达。结果:与正常组比较,模型组大鼠刻板行为和共济失调评分明显升高;其海马组织NRG1、ErbB4 mRNA的表达也明显升高。与模型组比较,温胆汤40、20、10 g/kg组均能明显降低大鼠刻板行为和共济失调评分;也能明显降低海马组织NRG1、ErbB4 mRNA的表达。结论:温胆汤通过降低精神分裂症易感基因NRG1及其受体ErbB4蛋白的表达,改善了大鼠的行为学改变,从而防治精神分裂症。
Objective: To research the effect of Wendan Tang on neuregulin-1 (NRG1) mRNA, ErbB4 mRNA of hippocampus and behavioral in rats with schizophrenia. Method: sixty SD rats were randomly divided six groups: normal group, model group, Clozapine 20 mg/kg group,Wendan Tang 40 g/kg group, Wendan Tang 20 g/kg group and Wendan Tang 10 g/kg group. The rats of normal group and model group were given normal saline, the rats of Clozapine 20 mg/kg group were given Clozapine 20 mg/kg, the rats of Wendan Tang 40, 20, 10 g/kg groups were respectively given Wendan Tang 40, 20, 10 g/kg, once a day, which last for 21 days. At the last given Wendan Tang, Clozapine or saline after two hours, except normal group, the rest of groups rats were ip given MK-801 0.6 mg/kg to establish rat schizophrenia model. After modeling, the behavioral changes of each group rats were observed and recorded. After 3 days,the rats were sacrificed, the hippocampus tissue were tested. To observe the behavioral changes, stereotyped behavior and ataxia score of rats. The expression of NRG1 mRNA and ErbB4 mRNA in rat hippocampus were detected by RT-PCR. Result: Compa/ed with the normal group, the stereotyped and ataxia scores of the model group rats were significantly increased ( P 〈 0.01 ), the expression of NRG1 mRNA and ErbB4 mRNA in hippocampus of the model group rats were also significantly increased( P 〈 0.05 ). Compared with the model group, Wendan Tang 40, 20, 10 g/kg groups rats could reduce the stereotyped and ataxia scores ( P 〈 0.05 ), and significantly reduce the expression of NRG1 mRNA and ErbtM mRNA in hippocampus( P 〈0.05 ). Conclusions: Wendan Tang could reduce the expression of NRG1 and its receptor ErbB4 as important risk genes for schizophrenia, improve behavioral changes in rats, which thus effectively preventing and treating schizophrenia.
