白豆蔻对阿霉素肾病模型大鼠肾脏组织TGF-β1、PAI-1表达的影响 被引量:7

Effect of Amomumkravanh Pierre ex Cagnep to expression of TGF-β1、PAI-1 on Adriamycin in duced renal injury Rats

摘要 目的:研究白豆蔻对阿霉素肾病大鼠的肾脏保护作用及其机制。方法:雄性Wistar大鼠60只,随机分为正常对照组、阿霉素肾病模型组、白豆蔻1.5 g/kg剂量组、白豆蔻3.0 g/kg剂量组、白豆蔻4.5 g/kg剂量组、尿毒清颗粒组。采用尾静脉一次性注射阿霉素(6.5mg/kg)建立阿霉素肾病模型,建立模型后第4d开始灌胃给药,连续给药3W。给药3W后代谢笼留取尿液,麻醉大鼠腹主动脉采血后处死动物并取肾脏组织,通过尿液分析仪检测各组大鼠尿液尿蛋白(PRO)、潜血(BLO)、胆红素(BIL)等指标;ELASE法检测血清Cys-C、尿液β2-MG的含量;免疫组化法检测肾脏组织TGF-β1、PAI-1的表达。结果:与模型组比较,各给药组均有不同程度的降低PRO、BLO、BIL含量,其中白豆蔻3.0 g/kg剂量组的作用优于其它给药组。ELASE法检测结果表明与模型组比较,白豆蔻三个剂量组均有降低Cys-C和β2-MG含量,其中白豆蔻3.0 g/kg剂量组和4.5 g/kg剂量组对Cys-C的作用优于低剂量组,白豆蔻3.0 g/kg剂量组对β2-MG的作用优于其它两个组;免疫组化实验结果表明与模型组比较肾组织TGF-β1、PAI-1的表达白豆蔻4.5 g/kg剂量组比较模型组明显减少,其中白豆蔻的各给药组对TGF-β1的表达没有明显区别。结论:白豆蔻能减轻阿霉素肾病大鼠的肾脏损伤,其作用机制可能与TGF-β1、PAI-1的表达有关。 Objective: To study mechanism and renal protective effects of Amomumkravanh Pierre ex Cagnep (AKP) against Adriamycin induced renal injury in Rats. Methods: Healthy 60 male Wistar rats, were randomly divided into normal group, model group, AKP1.5g/kg dose group, AKP 3.0g/kg dose group, AKP 4.5g/kg dose group, Niaoduqing granule drugs group. Induced renal injury using one time intravenous injection adriamyein (6.5mg o kg-1 ). establishment of the model After 4d ,administration for 3 consecutive weeks, after 3 weeks collect urine in metabolic cages. Anesthesia abdominal aorta of rats were sacrificed and kidney tissues were collected. Detected urine protein (PRO) , occult blood (BLO) , bilirubin (BIL) of each groups by urine analyzer; Detected the content of Cys-C in serum andβ2-MG in urine by ELASE; Observed pathological changes of renal tissue on light microscopy. Detected expression of PAI-1 (plasminogen activator enzyme) and LN (laminin) in renal tissues by immunohistochemistry. Results: compared with normal group ,the content of PRO,BLO,BIL in model group were increased significantly ; Compared with model group, content of PRO, BLO, BIL in each group increased in different level, the effect of AKP 3.0g/kg dose group is better than other groups. The result of ELASA detection showed that compared with normal group, content of Cys-C in rats serum andβ2-MG in urine were increased significantly; Compared with model group, AKP three dose groups were decreased content of Cys-C andβ2-MG,AKP 3.0g/kg dose group and 4.5g/kg dose group is better than 1.5g/kg does group,the effect of AKP 3.0g/kg close group onβ2-MG is better than other two does groups; HE staining showed that the Adriamycin induced renal damage group and Niaoduqing granule drugs group were havent significantly difference, disease was decreasing when increased dose of AKP three drug group ; The immunohistochemical results indicated that compared with model group,expression of TGF-β1 ,PAI-1 in renal tissues of AKP4.5g/kg dose group was decreased significantly,AKP each does group on expression of TGF-[31 was not significantly different. Conclusion: Amomumkravanh Pierre ex Cagnep (AKP) can decreased Adrianlycin induced renal injury, has a protective effect on kidney.
