剑叶耳草对ConA诱导的小鼠免疫性肝损伤的保护作用 被引量:3

摘要 目的:建立小鼠免疫性肝损伤模型,研究剑叶耳草水提物对小鼠免疫性肝损伤的保护作用。方法:采用刀豆蛋白A(Con A)尾静脉注射建立小鼠免疫性肝损伤模型,用比色分析法测定小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、总胆红素(TBIL)的活性;测定小鼠肝匀浆丙二醛(MDA)、还原型谷胱甘肽(GSH)的含量及谷胱甘肽过氧化物酶(GSH-Px)、过氧化物歧化酶(SOD)的活性;HE染色观察各组小鼠肝细胞损伤程度。结果:8g/kg和4g/kg剑叶耳草水提物能显著降低因Con A所致免疫性肝损伤小鼠血清异常升高的ALT、AST、ALP和TBIL;对免疫性肝损伤小鼠肝匀浆中MDA的含量明显降低,GSH含量明显升高及GSH-Px、SOD的活性有明显的升高作用。结论:剑叶耳草水提物对ConA诱导的免疫性肝损伤小鼠具有一定的保护作用,其机制可能与其抗氧自由基损伤、抑制脂质过氧化作用有关。 Objective: To establish the acute liver injury in mouse and observe the protective effect of Jianyeercao aqueous extract on acute liver injury in mice. Methods: The immunologic liver injury was induced by ConA injected into tail vein of mice. The calorimetric assay was used to measure the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) , alkaline phosphatase(ALP) and total bilirubin (TBIL) in mouse serum. And the content of malondialdehyde ( MDA), reduced glutathione (GSH) and activities of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) in rat liver homogenate were determined. Hematoxylineosin (HE) stain was used to examine the degree of hepatic injury. Results: 8g/kg and 4g/kg Jianyeercao aqueous extract obviously reduced the activities of AST, ALT, ALP and TBIL, and the content of MDA in immunologic hepatic injury mice, and increased the content of GSH and the activities of GSH-Px and SOD ( P 〈 0.01 or P 〈 0.05). Conclusion: Jianyeercao aqueous extract has an obvious protective effect on immunologic hepatic injury in mice. The mechanism may be related to attenuating free radical and inhibiting hpid peroxidation.
