摘要
目的:探讨栀子根醇提物对CCl_4致急性肝损伤小鼠的保护作用,为栀子根的临床应用提供实验依据。方法:腹腔注射0.15%的CCl_4花生油溶液诱导小鼠急性肝损伤,测定小鼠血清中ALT、AST活性,TBA、TBIL、DBIL水平,检测肝组织中SOD活性,TP、MDA水平,计算肝脏指数并观察小鼠肝脏病理学变化,采用多维尺度分析法综合评价栀子根醇提物对急性肝损伤小鼠的影响。结果:与模型组相比,栀子根醇提物(6g/kg,9g/kg)剂量组中ALT、AST、TBA、TBIL、DBIL、TP、SOD、MDA生化指标均有显著性改善,对小鼠肝组织病理性损伤有一定程度的减轻。结论:栀子根醇提物对CCl_4致急性肝损伤的小鼠有明显的保肝疗效,其机制可能与抗氧化作用、胆汁酸胆红素代谢的调节有关。
Objective: To provide scientific basis for the practical application of Radix Gardeniae, to study protection effects of the ethanolic extract of Radix Gardeniae on acute liver injury induced by CCl4 in mice. Methods:Acute liver injury model in mice was established by intraperitoheal injection of 0.15% CCl4 peanut oil solution. Activities of ALT,AST and contents of TBA,TBIL,DBIL in serum, activities of SOD and contents of TP, MDA in liver tissue were measured. Liver indexes and pathological changes of liver tissues were observed by microscope. Results:Compared with the model group, biochemical indexes of AST, ALT, TBA, TBIL, DBIL, TP, SOD and MDA were significantly improved in dose (6g/kg, 9g/kg) of the ethanolic extract group (P 〈 0.05). Pathological damage of liver tissues was eased to a certain degree. Conclusion:The ethanolic extract of Radix Gardeniae has protective effects on acute liver injury induced by CCl4 in mice. Its mechanism may be related to antioxidant effect and regulating metabolic functions of bile acid and bilirubin.
