摘要
本实验旨在研究务川黑牛SOCS基因家族的表达及调控,分别提取8头成年务川黑牛的背最长肌、腰大肌、肝脏、心脏、脾脏、小肠、肺、皮下脂肪共8个组织样品的RNA,应用实时荧光定量PCR技术检测务川黑牛SOCS基因家族在不同组织中m RNA的相对表达量。结果表明:务川黑牛SOCS基因家族成员m RNA在不同组织中均有表达,在脾脏和背最长肌中表达量较高,在腰大肌和心脏中表达量很低;CIS基因和SOCS2基因在多个组织中m RNA表达量较低且非常接近,CIS和SOCS2基因可能以低表达量形式协同参与生长调控;SOCS1和SOCS3在多个组织中m RNA表达量均较高且接近;SOCS4和SOCS5基因m RNA表达谱特征具有较高相似性,其功能发挥可能具有一定相关性。
The objectives of this study were to investigate the expression and regulation of the SOCS gene family in Wuchuan Black Cattle. The RNA of longissimus dorsi muscle, psoas muscle, liver, heart, spleen, small intestine, lung and subcutaneous fat of eight adult Wuchuan Black Cattles were extracted, and real-time quantitative PCR method was used to detect the relative expression amount of SOCS gene family in different tissues in Wuchuan Black Cattle. The results showed that the mRNA of SOCS gene family members had the expression in all tissues, and the higher expression was in spleen and longissimus dorsi muscle, while the lower expression was in psoas muscle and heart. The mRNA of CIS and SOCS2 gene had lower and similar expression in various tissues, which indicated that CIS and SOCS2 gene might synergistically regulate the growth with low expression form. However, the mRNA of SOCSI and SOCS3 gene in several tissues was higher and similar. The mRNA expression profiles of SOCS4 and SOCS5 gene were quite similar, therefore their function might have some relevance.
