摘要
目的探讨绝经后女性股骨头坏死的发生与血清miR-1207-5p和miR-96-5p表达水平的关系。方法连续选择30例绝经后女性股骨头坏死患者(观察组)和30例绝经后女性健康志愿者(对照组),采用反转录PCR(RT-PCR)法检测外周循环miR-1207-5p和miR-96-5p表达水平,ELISA法检测循环骨形成蛋白(BMP)-2和血管内皮生长因子(VEGF)表达水平。结果观察组血清miR-1207-5p表达水平明显高于对照组,miR-96-5p表达水平低于对照组,miR-1207-5p上调率和miR-96-5p下调率均明显高于对照组,差异有统计学意义(P<0.05)。观察组血清BMP-2和VEGF表达水平均明显低于对照组,差异有统计学意义(P<0.05)。结论 miR-1207-5p和miR-96-5p表达异常可能与绝经后女性股骨头坏死的发生有关,影响BMP-2和VEGF的表达水平可能是其发生机制。
Objective To explore the relationships between expression levels of serum miR-1207-5p and miR-96-5p and femoral head necrosis of postmenopausal women. Methods Thirty postmenopausal women with femoral head necrosis ( observation group) and thirty healthy postmenopausal women (control group) were selected, RT-PCR was used to detect the expression levels of miR-1207-5p and miR- 96-5p in peripheral blood. ELISA was used to detect the expression levels of bone morphogenetic protein-2 (BMP-2) and vascular endo- thelial growth factor (VEGF) . Results The expression level of serum miR-1207-5p in observation group was higher than that in control group, while the expression level of serum miR-96-5p in observation group was lower than that in control group, the upregulation rate of miR-1207-5p and downregulation rate of miR-96-5p in observation group were higher than those in control group, there were statistically significant differences (P〈0. 05 ) . The expression levels of serum BMP-2 and VEGF in observation group were statistically significantly lower than those in control group (P〈0.05) . Conclusion The abnormal expressions of miR-1207-5p and miR-96-5p may be correlated with femoral head necrosis of postmenopausal women, the effects on expression levels of BMP-2 and VEGF are the probable mechanisms.
