摘要
目的探讨Yes相关蛋白(YAP)、细胞周期蛋白D1(Cyclin D1)在宫颈癌前病变与宫颈鳞癌中的表达。方法收集100例患者宫颈病变组织,其中宫颈鳞癌35例、CIN48例、正常宫颈组织17例,以免疫组织化学法检测以上组织标本中YAP、Cyclin D1的表达。结果宫颈鳞癌组织、宫颈上皮内瘤变(CIN)组织和正常宫颈组织中YAP阳性表达率分别为65.71%、41.67%、11.76%,Cyclin D1阳性表达率分别为60.00%、33.33%、0.00%,阳性表达率在不同组织中比较差异有统计学意义(P<0.05),YAP、Cyclin D1阳性表达率宫颈鳞癌组织均高于CIN组织及正常宫颈组织(P<0.05),CIN组织高于正常宫颈组织(P<0.05);宫颈鳞癌临床分期III-IVYAP、Cyclin D1阳性表达率为80.00%、75.00%高于Ⅰ~Ⅱ期者46.67%、40.00%(P<0.05),有淋巴结转移者YAP、Cyclin D1阳性表达率为78.26%、73.91%高于Ⅰ~Ⅱ期者41.67%、33.33%(P<0.05);CINⅢ期者YAP、Cyclin D1阳性表达率为55.56%、48.15%高于Ⅱ期者23.81%、14.29%(P<0.05)。结论 YAP、Cyclin D1在宫颈癌前病变与宫颈鳞癌中的表达均有上调,并与病情发生及发展密切相关。
Objective To explore the expressions of Yes-associated protein( YAP) and Cyclin D1 in cervical intraepithelial neoplasia and cervical squamous cell carcinoma. Methods A total of 100 cases with cervical lesions were collected,including 35 cases of cervical squamous cell carcinoma,48 cases of cervical intraepithelial neoplasias( CIN),and 17 cases of normal cervical tissue. Immunohistochemical method was used to detect the expressions of YAP and Cyclin D1. Results The positive expression rates of YAP in cervical squamous cell carcinoma,CIN,and normal cervical tissue were 65. 71%,41. 67%,and 11. 76%,respectively. The positive expression rates of Cyclin D1 in cervical squamous cell carcinoma,CIN,and normal cervical tissue were 60. 00%,33. 33%,and 0. 00%,respectively. There were statistically significant differences in positive expression rates of YAP and Cyclin D1 among different cervical tissue samples( P 0. 05). The positive expression rates of YAP and Cyclin D1 in cervical squamous cell carcinoma were statistically significantly higher than those in CIN and normal cervical tissues( P〈0. 05). The positive expression rates of YAP and Cyclin D1 in CIN were statistically significantly higher than those in normal cervical tissue( P〈0. 05). The positive expression rates of YAP and Cyclin D1 in cervical squamous cell carcinoma of clinical stage Ⅲ-Ⅳ were 80. 00% and 75. 00%,respectively,which were statistically significantly higher than those in cervical squamous cell carcinoma of clinical stage Ⅰ-Ⅱ( 46. 67% and 40. 00%,respectively)( P 0. 05). The positive expression rates of YAP and Cyclin D1 in cervical squamous cell carcinoma patients with lymph node metastasis were 78. 26% and 73. 91%,respectively,which were statistically significantly higher than those in cervical squamous cell carcinoma patients without lymph node metastasis( 41. 67%and 33. 33%,respectively)( P〉 0. 05). The positive expression rates of YAP and Cyclin D1 in CIN of stage Ⅲ were 55. 56% and48. 15%,respectively,which were statistically significantly higher than those in CIN of stage Ⅱ( 23. 81% and 14. 29%,respectively)( P〈0. 05). Conclusion The expressions of YAP and Cyclin D1 in CIN and cervical squamous cell carcinoma are up-regulated,which are closely related to occurrence and development of the disease.
