人参和三七HPLC特征图谱研究 被引量:4

Study on HPLC Characteristic Fingerprint of Panax Ginseng Radix C.A.Mey. and Panax notoginseng(Burk.) F.H.Chen

摘要 目的:建立人参与三七的高效液相色谱法(HPLC)特征图谱分析方法 ,区别人参和三七药材粉末。方法 :采用HPLC法,色谱柱:Kromasil-C18柱(4.6 mm×250 mm,5μm),流动相:乙腈-水(0.1%磷酸),梯度洗脱,流速0.9 m L·min-1,柱温25℃,检测波长203 nm,分析时间75 min。采用"中药色谱指纹图谱相似度评价系统(2004A版)"进行相似度分析;采用Matlab 7.0和SPSS 16.0软件进行主成分分析和聚类分析。结果 :分别得到人参和三七的HPLC特征图谱及共有模式图,通过对比,两者共有峰为16个,分别为a^p,并指认了其中的人参皂苷Rg1、人参皂苷Re、人参皂苷Rf、人参皂苷Rb1、人参皂苷Rd 5个共有峰;人参中的特征峰为1~9,三七中的特征峰为10~32;相似度分析结果显示,人参的相似度值在0.941~0.992,三七的相似度值在0.755~0.963;主成分分析和聚类分析将供试药材分为两大类。结论:该分析方法可以快速简便地区别人参和三七药材粉末,并为人参和三七药材的质量控制标准制定提供了依据。 Objective:To develop the HPLC specific chromatogram of Panax ginseng Radix C. A. Mey. and Panax notoginseng(Burk.) F. H. Chen for distinguishing between their powders. Methods:A HPLC analysis was carried out on a Kromasil Cls column(4.6 mm×250 mm,5 μm) with the mobile phase consisting of acetonitrile(A)-water(0.1% phosphoric acid) in gradient mode. The flow rate was 0.9 mL·min^-1 and the col- umn temperature was kept at 25℃. The detection wavelength was set at 203 nm and the analytic time was 75 min. Similarity evaluation system for chromatographic fingerprint of TCM(2004A) was used to conduct similarity evaluation and Matlab 7.0 and SPSS 16.0 softwares were used for principal component and cluster analysis. Results:Panax Ginseng Radix C. A. Mey. and Panax Notoginseng(Burk.) F. H. Chen had sixteen common peaks,namely a - p,five common peaks were identified,ginsenoside-Rg1,ginsenoside-Re, ginsenoside-Rf,ginsenoside-Rbl and ginsenoside-Rd respectively. The characteristic peaks of ginseng were 1-9, while the characteristic peaks of notoginseng were 10-32. Similarity value of Panax ginseng Radix C. A. Mey. and Panax notoginseng(Burk.) F. H. Chert were 0.941-0.992 and 0.755-0.963. According to the principal component analysis and clustering analysis,the tested samples were classified into two categories. Conclusion:The method can be used to distinguish between ginseng and notoginseng powder quickly and easily and provides new evidence for the quality control of Panax ginseng Radix C. A. Mey. and Panax notoginseng(Burk.) F. H. Chen.
