摘要
目的:建立同时测定维生素AD小丸中维生素A棕榈酸酯和维生素D_2含量的反相高效液相色谱法。方法:采用Phenomenex C_(18)色谱柱,甲醇为流动相,流速1.0 mL·min^(-1),柱温35℃,检测波长265 nm,进样量10μL,按内标法计算维生素A棕榈酸酯和维生素D_2的含量。结果:维生素A棕榈酸酯和维生素D_2线性范围分别为522.3~208 9.4 IU·mL^(-1)(r=0.999 9)和51.88~207.5 IU·mL^(-1)(r=0.999 9),平均回收率分别为100.3%和103.4%,定量限分别为1.979 IU·mL^(-1)和3.179 IU·mL^(-1)。结论:本方法操作简单、快速、准确,可同时测定维生素AD微珠中维生素A棕榈酸酯和维生素D_2的含量。
Objective:To establish reversed-phase high performance liquid chromatography (RP-HPLC) methods for determination of Vitamin A Palmitate and Vitamin D2 in Vitamin AD Beads. Method: The determination was car- ried out on a phenomenex CIS column. The mobile phase was methanol. The flow rate was 1.0 mL ·min^-1, the column temperature was 35 ℃, the detection wavelength was 265 nm and the injection volumn was 10 μL. Vitamin A Palmitate and Vitamin D2 were calculated by internal standard method. Result: The good linearity of Vitamin A Palmitate and Vitamin D2 were 522.3 - 208 9.4 IU ·mL^-1 ( r = 0. 999 9) and 51.88 - 207.5 IU ·mL^-1 ( r =0. 999 9), the average recoveries were 100.3% and 103.4% , the LLOQ were 1. 979 and 3. 179 IU ·mL^-1 respectively. Conclusion : The method is simple, rapid and accurate, which can be used for the determination of Vitamin A Pal-mitate and Vitamin D2 in Vitamin AD Beads.
