The Effect of Neutrophil Extracellular Traps on the Human Monocytes Proinflammatory Cytokines Secretion

摘要 目的:探讨中性粒细胞胞外诱捕网(NETs)对单核细胞分泌炎症介质的影响。方法:采用健康人中性粒细胞体外诱导NETs形成,用该NETs刺激单核细胞,LEGENDplex人炎症因子试剂盒检测培养上清多种炎症介质的变化。结果:单核细胞与NETs共培养12 h后,NETs组与对照组相比,IL-1β诱生量分别为(97.60±60.11)pg/ml、<1.17 pg/ml,IFN-α诱生量分别为(208.90±77.38)pg/ml、<1.17 pg/ml,TNF-α诱生量分别为(218.88±86.27)pg/ml、<0.17 pg/ml,MCP-1诱生量分别为(1 063.25±740.01)pg/ml、(157.16±3.86)pg/ml,IL-6诱生量分别为(67.76±15.51)pg/ml、(2.76±0.47)pg/ml,差异均具有统计学意义(P<0.05),IL-10、IL-12表达水平差异无统计学意义。结论:NETs能显著促进单核细胞分泌IL-1β、IFN-α、TNF-α、MCP-1、IL-6。 Objective:Oevaluate the effect of neutrophil extracellular traps (NETs) on the human monocytes proinflammationtory cytokines release. Methods:Neutrophils isolated from healthy human peripheral blood were stimulated with phorbol myristate acetate (PMA)to induce NETs production. After stimulated with NETs, the concentrations of interleukin - 1β (IL - 1β), interferon - α ( IFN -α), tumor necrosis factor - α( TNF -α), monocyte chemoattractant protein - 1 ( MCP - 1 ), interleukin - 6 ( IL - 6 ), interleukin- 10 (IL- 10) and interleukin- 12 (IL- 12) in supernatant were analyzed using LEGENDplex reagents. Results:After twelve hours' culture, the proinflammatory cytokines' secretion of monocytes stimulated by NETs was significantly higher than that non - stimulated by NETs, including IL - 113 [ (97.60 ± 60.11 ) pg/ml vs 〈 1.17 pg/ml ], IFN - α[ ( 208.90± 77.38 ) pg/ml vs 〈1.17pg/ml], WNF-α[(218.88±86.27)pg/mlvs 〈0.17pg/ml], MCP-1[(1 063.25±740.01)pg/mlvs(157.16±3.86) pg/ml] and IL -6 [ (67.76± 15.51) pg/ml vs 2.76 ±0.47) pg/ml] ,P 〈0.05. IL - 10 and IL - 12 levels in both groups had no significant difference. Conclusion:NETs can significantly increase proinflammationtory eytokines' secretion of monocytes, including IL - 1β, IFN - α, TNF -α, MCP - 1 and IL - 6.
