摘要
目的:对三七姜挥发油成分进行定性和定量分析并探讨挥发油对不同肿瘤细胞株的生长抑制作用。方法:采用水蒸气蒸馏法测定三七姜挥发油的含量,并用气相色谱-质谱(GC-MS)计算机联用技术分离鉴定其中的挥发性化学组成,用归一化法测定其百分含量。采用噻唑蓝(MTT)比色法检测三七姜挥发油对体外培养的人鼻咽癌细胞系CNE-2细胞株、人肺癌细胞系A549细胞株、人肝癌细胞系Hep G2细胞株、人乳腺癌细胞系MCF-7细胞株的生长抑制作用,计算半数抑制浓度(IC50)。结果:已鉴定挥发油成分占总挥发油含量为94.03%,各成分分离度较好,初步鉴定了58种成分,其中含9类化合物,单萜类化合物,单萜氧化物类化合物,倍半萜类化合物,倍半萜氧化物类化合物,酯类化合物,芳烃类化合物,醛类化合物,醇类化合物和烷烃类化合物。含量最高的是3,6,7,8-四氢化-3,3,6,6-四甲基环戊二烯并[E]茚-1(2H)-酮,占25.75%;其次是莰烯,占13.54%;居第3位的是1,7,7-三甲基-二环[2.2.1]庚-2-茨酮,占11.93%。三七姜挥发油能抑制不同肿瘤细胞(CNE-2、A549、Hep G2、MCF-7)的增殖,且呈剂量依赖性,IC50值分别为1.27,1.24,0.84和1.04 mg/ml。结论:三七姜挥发油具有体外抗肿瘤活性。以上研究为三七姜挥发油进一步开发利用提供实验基础,为寻找三七姜中抗肿瘤的有效组分或新的化合物提供科学依据,抗肿瘤活性有待进一步研究。
Objective: To make qualitative and quantitative analysis of components of essential oil of stahlianthus involucratus and investigate its growth inhibition mechanism in different tumor cell lines.Methods: Steam distillation method was used to detect the contents of essential oil of stahlianthus involucratus,GC-MS computer combined technology was applied to separating and determinating its chemical components,and normalization method was used to determine its percentage content. Then MTT assay was used to determined the growth inhibition effect of essential oil of stahlianthus involucratus on invitro cultured CNE-2 cell line,A549 cell line,Hep G2 cell line,and Mc F-7 cell line,and half of the inhibition concentration( IC50) was calculated. Results: The essential oil of stahlianthus involucratus was identified as 94. 03% of the total contents with better separation. 58 components had been identified,including 9 classes of chemical compounds: monoterpenes, monoterpene oxide compounds,sesquiterpenoids,sesquiterpenoids oxide compounds,ester compounds,aromatic compound,aldehyde chemical compound,Sterols and alkanes compounds. The highest content is 3,6,7,8-tetrahydrofuran polymers-3,3,6,6-tetramethyl cyclopentadiene and [E] indene-ketone,accounting for 25.75%; followed by camphene,accounting for 13. 54%, the third is 1,7,7-trimethylbicyclo [2. 2. 1] heptan-2-one,accounting for 11. 93%. The essential oil can inhibit the proliferation of different tumor cells( CNE-2,A549,Hep G2,MCF-7),which are dose-dependent,and the IC50 value are 1. 27,1. 24,0. 84 and1.04mg/ml respectively. Conclusion: The essential oil of stahlianthus involucratus has in-vitro antitumor activity. The study may provide experimental basis for the development and application of the essential oil,and also provide scientific basis for the development of effective antitumor components or the new compounds,and the antitumor activity needs further study.
