摘要
目的探讨磷脂酰肌醇-3-激酶/蛋白激酶B(PI3K/Akt)通路在子宫内膜腺癌上皮间质转化(EMT)过程中的作用。方法采用免疫组化PV-6000法检测12例正常增殖期子宫内膜、16例子宫内膜非典型增生、32例子宫内膜癌组织中磷酸化Akt(p-Akt)、上皮-钙黏蛋白(E-cadherin)及波形蛋白(Vimentin)的表达水平,并分析三者间及与各项临床病理参数的关系。结果 3组内膜组织中p-Akt、E-cadherin和Vimentin阳性表达率比较,差异有显著性(χ2=13.91~19.28,P<0.05)。且在子宫内膜癌组织中p-Akt蛋白的表达与肿瘤病理分期及淋巴结转移有关(P=0.041、0.047),与肌层浸润程度和组织学分级无关(P=0.676、0.070);E-cadherin蛋白的表达与病理分期、肌层浸润程度和淋巴结转移有关(P=0.008、0.029、0.026),与组织学分级无关(P=0.640);Vimentin蛋白的表达与病理分期及淋巴结转移有关(P=0.019、0.012),与肌层浸润程度和组织学分级无关(P=0.647、0.648)。子宫内膜癌组织中p-Akt与Vimentin的表达水平呈正相关(r=0.462,P<0.05),与E-cadherin的表达水平呈负相关(r=-0.567,P<0.05)。结论 PI3K/Akt通路可能通过介导EMT参与子宫内膜癌的浸润与转移。
Objective To examine the role of PI3K/Akt signaling pathway in the process of epithelial-mesenchymal transition(EMT)in endometrial adenocarcinoma. Methods The protein expression of p-Akt,E-cadherin,and vimentin in 12 normal endometrium tissues,16 endometrium hyperplasia tissues,and 32 endometrial adenocarcinoma tissues was determined by PV-6000 immunohistochemistry.The relationship between the three proteins and each clinicopathological parameter was analyzed.Results The rates of p-Akt,E-cadherin,and vimentin expression were significantly different between the three groups(χ2=13.91-19.28,P〈0.05).The expression of p-Akt in endometrial carcinoma tissues was associated with pathological stage and lymph node metastasis(P=0.041 and 0.047,respectively),but not with the degree of myometrial invasion and histological grade(P=0.676 and 0.070,respectively).The expression of E-cadherin was associated with pathological stage,degree of myometrial invasion,and lymph node metastasis(P=0.008,0.029,and 0.026,respectively),but not with histological grade(P=0.640).The expression of vimentin was associated with pathological stage and lymph node metastasis(P=0.019 and 0.012,respectively),but not with degree of myometrial invasion and histological grade(P=0.647 and 0.648,respectively).In addition,the expression of p-Akt was positively correlated with vimentin expression(r=0.462,P〈0.05),and was negatively correlated with E-cadherin expression in endometrial carcinoma tissues(r=-0.567,P〈0.05). Conclusion The PI3K/Akt signaling pathway may be involved in tumor invasion and metastasis through mediating the EMT of endometrial adenocarcinoma.
