摘要
目的探讨硅沉着病患者血清及肺组织中载脂蛋白A、基质金属酶-9、组织蛋白酶G的表达情况。方法选择2010年2月—2016年1月广元市第一人民医院收治的80例硅沉着病患者为硅沉着病组,因胸部外伤行肺部手术治疗的39例为对照组,采用酶联免疫吸附法检测血清中载脂蛋白A(Apo)、基质金属酶-9(MMP-9)、组织蛋白酶G水平,用组织化学免疫检测肺组织中载脂蛋白A、基质金属酶-9、组织蛋白酶G表达,logistic回归性分析三者与硅沉着病的相关性。结果硅沉着病组血清中的基质金属酶-9和组织蛋白酶G水平明显高于对照组[(140.56±20.17)vs(228.33±35.09)μg/L;(3.18±0.53)vs(7.92±1.31)ng/ml],载脂蛋白A水平低于对照组[(203.31±29.86)vs(44.36±8.01)μg/ml],差异有统计学意义(t=21.37,11.75,9.83,P<0.05);硅沉着病患者肺组织中基质金属酶-9和组织蛋白酶G表达呈强阳性,载脂蛋白A在正常肺组织中高表达,在硅沉着病患者中低表达,而正常肺组织中两者几乎不表达。Logistic回归性分析提示,载脂蛋白A、基质金属酶-9和组织蛋白酶G升高均是硅沉着病的危险因素(P<0.05)。结论载脂蛋白A、基质金属酶-9、组织蛋白酶G在硅沉着病患者肺组织中高表达,可能是硅沉着病潜在的生物学标志物。
[Objective] To explore the expression of apolipoprotein A,matrix metalloproteinase-9 and cathepsin G in serum and lung tissue of patients with silicosis.[Methods] 80 silicosis patients who were treated in the First People's Hospital of Guangyuan City from February 2010 to January 2016 were enrolled in the silicosis group,while patients undergoing pulmonary surgery due to thoracic trauma were recruited in the control group.The level of apolipoprotein A,matrix metalloproteinase-9 and cathepsin G in serum were detected by enzyme-linked immunosorbent assay,and the expression of apolipoprotein A,matrix metalloproteinase-9 and cathepsin G in lung tissue were detected by histochemical immunoassay.Logistic regression method was used to analyze the correlation between them and silicosis.[Results] The serum levels of matrix metalloproteinase-9 and cathepsin G in the silicosis group were significantly higher than those in the control group [(140.56 + 20.17) vs (228.33 + 35.09) μg/l,(3.18 + 0.53) vs (7.92 + 1.31) ng/ml],the serum level of apolipoprotein A in the silicosis group was lower than that in the control group [(203.31 &#177;29.86) vs (44.36 &#177;8.01) μg/ml],and the differences were statistically significant (t =21.37,11.75,9.83,P〈0.05).The expressions of matrix metalloproteinases-9 and cathepsin G were strongly positive in patients with silicosis,while those almost had no expression in normal lung tissue.The expression of apolipoprotein A was strongly positive in normal lung tissue,while that was low in patients with silicosis.Logistic regression analysis showed that elevated levels of apolipoprotein A,matrix metalloproteinase-9 and cathepsin G were risk factors of silicosis (P〈0.05).[Conclusion]The apolipoprotein A,matrix metalloproteinase-9 and cathepsin G express highly in lung tissue of patients with silicosis,which may be the potential biomarkers of silicosis.
