摘要 ‘青砧D1’是青岛农业大学选育的一个梨矮化砧优系。本试验研究了不同浓度的TDZ、6-BA、IBA及IAA配比对‘青砧D1’叶片离体再生培养的影响,从而为该砧木的快速繁殖和应用推广奠定基础。结果表明:MS培养基中附加适宜浓度和配比的6-BA与IBA能提高初代培养外植体的成活率,以6-BA 1.0mg·L^(-1)与IBA 0.1mg·L^(-1)浓度配比的外植体成活率最高,达86.3%。NN69培养基中附加不同浓度的TDZ与IBA或IAA能有效地诱导叶片外植体不定芽再生,以TDZ和IBA配合诱导产生不定芽的外植体比例高,在TDZ 4.0mg·L^(-1)与IBA 0.3mg·L^(-1)配比组合下可达到100%,但平均外植体再生芽数为3个,低于TDZ与IAA配比组合(3.9个/外植体)。在所试的不同浓度TDZ与IAA配比组合中,外植体不定芽诱导率最高仅为68%。MS培养基中附加6-BA 1.0mg·L^(-1)和IBA 0.2mg·L^(-1)时芽增殖系数最高,达4.42。在1/2 MS培养基中附加IBA 0.3mg·L^(-1)时生根率最高,达87.3%,移栽成活率达95%。 ' Qing zhen DI' was a excellent pear dwarfing rootstock line selected by Qingdao Agricultural U- niversity. In this experiment, the effects of different combinations and concentrations of cytokinin (TDZ, 6-BA) and auxin (IBA, IAA) on the regeneration were examined in 'Qing zhen DI~, laying the foundation for rapid propagation, application and promotion of this rootstock. The results showed that MS supple- mented with appropriate concentrations of 6-BA and IBA could improve the survival rate of primary cul- tured explants, up to 86.3% with 6-BA 1.0 mg· L^-1+ IBA 0.1 mg· L^- 1. Combination of TDZ and IBA or IAA on NN69 medium could effectively induce the adventitious bud. And the relatively high regenera- tion rate was induced by TDZ and IBA, could up to 100%, with TDZ 4.0 mg · L^-1+ IBA 0.3 mg ·L-1. But the average induced adventitious bud number was just 3 per explant, which was lower than that of TDZ and IAA combination (3.9/explant). Among the different concentration combinations of TDZ and IAA tested, the hightest induction rate of adventitious bud was only 68 %. MS medium supplemented with 6-BA 1.0 mg · L-1 and IBA 0.2 mg · L-1 could induce the highest bud multiplication coefficient, up to 4:. 42. The hightest rooting rate was 87.3% and the survival rate of transplanting was 95% when 0.3 mg ·L-1 IBA was added in 1/2 MS medium.
