摘要
目的:探讨三黄益肾胶囊对糖尿病大鼠肾组织NO及NOS的影响。方法:采用链脲佐菌素(STZ)诱导糖尿病模型,随机分为糖尿病模型组、三黄益肾胶囊低、中、高剂量治疗组,每组10只,取10只正常大鼠为正常对照组。第8周末测定血糖、血肌酐、尿素氮、肾质/体质量比、24 h尿蛋白定量,测定干预后各组大鼠肾组织匀浆NO和NOS含量。结果:三黄益肾胶囊低、中、高剂量组大鼠血糖与模型组比较,差异均无统计学意义(P>0.05);三黄益肾胶囊低、中、高剂量组大鼠血肌酐、尿素氮、肾质/体质量和24 h尿蛋白定量明显低于模型组(P<0.05)。三黄益肾胶囊低、中、高剂量组、模型组大鼠肾脏组织匀浆NO和NOS均低于正常对照组(P<0.05);三黄益肾胶囊中、高剂量组大鼠肾脏组织匀浆NO和NOS均高于模型组(P<0.05)。结论:三黄益肾胶囊可降低糖尿病大鼠血肌酐、尿素氮及24 h尿蛋白水平,升高糖尿病大鼠肾组织匀浆NO及NOS含量,对糖尿病肾病具有保护作用。
Objective: To investigate the effect of Sanhuang Yishen capsule (三黄益肾胶囊, SHYSC) on NO and NOS of kidney tissues in diabetic rats. Methods: The diabetic model rats which were induced by STZ were randomly divided into diabetic model group, SHYSC high, medium and low dose treatment groups, 10 rats in each group. And get 10 normal rats in normal control group. After 8 weeks, blood glucose, serum creatinine, blood urea nitrogen, kidney weight/body weight ratio, 24 h urine protein quantitative determination, NO and NOS of kidney tissue were determined. Results: Compared with the model group, the difference of the blood glucose between the SHYSC low, middle and high dose groups was not statistically significant (P〉0.05). The serum creatinine, urea nitrogen, renal matter/body mass and 24 h urinary protein in the SHYSC low, middle and high dose groups were significantly lower than those in the model group (P〈0.05). The levels of NO and NOS in the kidney tissue homogenate of the SHYSC low, middle and high dose group and model group were lower than that of the normal control group (P〈0.05); The levels of NO and NOS in the kidney homogenate of the middle and high dose group were higher than that of the model group (P〈0.05). Conclusion: SHYSC can reduce serum creatinine, blood urea nitrogen and 24h urine protein, increase NO and NOS content of diabetes rats renal tissue, has a protective effect on diabetic nephropathy.
