两种Real-Time PCR试剂在甲乙型流感检测中的应用

Application of two kinds of Real-time PCR kit for detection of influenza A/B Virus

目的评价不同实时荧光定量PCR检测试剂盒应用于甲乙型流感病毒检测的可行性。方法两种试剂对已知结果的咽拭子标本进行平行检测,比较检测结果的特异性、灵敏度、符合率以及定量检测结果的差异。结果不同试剂甲乙型流感检测特异性均为100%,试剂a检测甲乙型流感的灵敏度分别为97.78%和96.67%,符合率分别为98.83%和97.78%;试剂b检测甲乙型流感的灵敏度均为93.33%,符合率分别为95.00%和95.56%,两种试剂盒检测甲乙型流感Ct值差异均有统计学意义(t值分别为-16.27和-2.19,P<0.05)。结论试剂a总性能优于试剂b,更加适用于作为流感监测的检测试剂。

Objective To evaluate the feasibility of application of different kinds of Real-time PCR kit for detection of influenza A/B viral. Methods Two kinds of kits were used for parallel detection of throat swab samples of known result,comparing the specificity,sensitivity,coincidence rate of different kits and the difference of quantitative results. Results The specificities of influenza A and B were both 100%,The sensitivities of influenza A and B by kit a were 97. 78% and 96. 67%respectively,the coincidence rates were 98. 83% and 97. 78% respectively; The sensitivities of influenza A and B by kit b were both 93. 33%,the coincidence rates were 95. 00% and 95. 56% respectively. There was significant difference between the two reagents in the quantitative detection of influenza A and B（ Ct）. Conclusion The total function of kit a is better than that of the kit b,which is more suitable for the detection kit for the primary screening of influenza monitoring.
