摘要
目的观察人肿瘤坏死因子凋亡相关蛋白(hTRAIL)对大鼠胶原诱导的类风湿关节炎模型(CIA)滑膜增殖作用的影响。方法 Wistar雄性大鼠随机分成6组:空白对照组、空白给药(hTRAIL 1μg)组、模型组、hTRAIL 1μg组、hTRAIL 1.5μg组、hTRAIL 2μg组。建立大鼠CIA模型,计量方法测定大鼠踝关节周径及其肿胀率,HE染色法观察关节病理形态学变化,Tunel法检测滑膜成纤维细胞凋亡情况,免疫组化法检测DR5表达情况。结果 hTRAIL干预模型组踝关节周径肿胀率较模型组低,关节破坏较模型组轻,滑膜细胞凋亡较模型组明显,DR5表达情况较模型组高(P<0.05),并呈剂量依赖性。结论 hTRAIL可促进CIA大鼠滑膜细胞的凋亡,减轻CIA大鼠的关节肿胀及诱导DR5表达水平升高。
Objective To investigate the inhibitory effect of hTRAIL on synovial proliferation in rats with collagen induced arthritis. Methods 36 wistar rats were randomly divided into six groups, the control ( groupl ), control 1 ixg hTRAIL - treated ( group 2 ), model group ( group 3 ), 1 p,g hTRAIL - treated ( group 4 ), 1.5 μg hTRAIL - treated group ( group 5 ), 2 μg hTRAIL - treated ( group 6 ). CIA model were created. Ankle joint diameter and swelling rate were adopted to evaluate the inflammatory reaction in rats. HE staining was used to observe the joint pathological morphology. The synovieytes apoptosis was detected by the tunel. DR5 expression was detected by the immunohistochemistry. Results Compared with the model group, the Ankle joint swelling rate was lower and joint damage was lighter, synovial cell apoptosis was more ob- vious and DR5 expression was higher in the hTRAIL - treated model groups. The difference was statistically significant ( P 〈 0.05 ). Conclusion hTRAIL can enhance synovicytes apoptosis, reduce the joint swelling and increase the expression level of DtL5 in rats with collagen induced arthritis.
