摘要
目的探讨神经突触核蛋白-γ(SNCG)、骨形态发生蛋白2(BMP-2)、基质金属蛋白酶9(MMP-9)在口腔颌面部鳞癌中的表达情况及相关性。方法选取2010年1月至2015年12月存档的口腔颌面部鳞癌组织蜡块标本67例,以及相应的癌旁组织67例和41例正常组织作为对照组。采用免疫组化En Vision二步法检测SNCG、BMP-2、MMP-9在三组的表达,并分析SNCG、BMP-2、MMP-9三者之间的相关性。结果 SNCG和MMP-9在肿瘤组织中阳性率显著高于在正常组织和癌旁组织(P<0.05),而SNCG和MMP-9在癌旁组织与正常组织中的阳性率比较差异无统计学意义(P>0.05);BMP-2在肿瘤组织和癌旁组织中的阳性率显著高于正常组织(P<0.05),但BMP-2在肿瘤组织和癌旁组织中的阳性率比较后差异无统计学意义(P>0.05)。BMP-2、MMP-9中在病理分级中的阳性率比较差异无统计学意义(P>0.05),SNCG在高分化组的阳性表达率显著低于在中、低分化组(P<0.05),而中、低分化组中SNCG的表达差异无统计学意义(P>0.05)。除BMP-2外,SNCG、MMP-9均在已发生淋巴结转移的组织中的阳性表达率显著高于在未发生淋巴结转移的组织(P<0.05)。SNCG、BMP-2、MMP-9三者的表达均呈正相关(P<0.05)。结论 SNCG、BMP-2、MMP-9在口腔颌面部鳞癌中呈高表达,且三者呈显著正相关,提示三者可以作为检测口腔颌面部鳞癌的生物学指标。
Objective To discuss the correlation between synuclein gamma( SNCG),bone morphogenetic proteins-2( BMP-2),matrix metalloproteinase-9( MMP-9) and the clinicopathological features of tumor,the expression of SNCG,BMP-2 and MMP-9 in oral and maxillofacial carcinoma and its adjacent tissues and normal tisssues were investigated. Methods Immunohistochemistry method was used to detect the expression of SNCG,BMP-2 and MMP-9 in 67 cases of oral and maxillofacial carcinoma,67 cases of its adjacent tissues and 41 cases of normal tisssues. Results The incidence rates of SNCG and MMP-9 in tumor tissues were significantly higher than in normal tissues and cancer adjacent tissues( P〈0. 05). While the incidence rates of SNCG and MMP-9 in the adjacent tissue and normal tissues had no statistically significant difference( P〈0. 05). The incidence rates of BMP-2 in tumor tissue and adjacent tissue were significantly higher than that in normal tissue( P〈0. 05). But it had no statistically significant difference( P〈0. 05) in tumor tissue and tissue adjacent. The positive rates of BMP-2,MMP-9 had no statistical significance( P〈0. 05) in pathological grading; the positive expression rate of SNCG in high differentiation group was significantly lower than in the middle and low differentiation groups( P〈0. 05),while in middle and low differentiation groups the expressions of SNCG had no significant difference( P〉0. 05). Except for BMP-2,the expression of SNCG and MMP-9 in metastatic tumor were higher than that in non-metastatic tumor,and there was significant difference( P〈0. 05). The expression of SNCG,BMP-2 and MMP-9were positively correlated( P〈0. 05). Conclusion The excessive expression of SNCG,BMP-2 and MMP-9 in oral and maxillofacial carcinoma is positively associated with the differentiation degree of carcinoma. So the expression of SNCG,BMP-2 and MMP-9 are expected to be the marker in the tumor of oral and maxillofacial carcinoma for the judgment.
