生长抑制因子2在人食管鳞癌组织的表达及其临床意义

Expression of growth inhibitory factor 2 in human esophageal squamous cell carcinoma and clinical significance

目的探讨人食管鳞癌中生长抑制因子2（ING2）的表达及其临床意义。方法应用免疫组织化学检测63例食管鳞癌和30例癌旁组织中ING2蛋白表达水平，实时定量反转录聚合酶链反应（RT-qPCR）检测19例食管鳞癌组织和17例正常食管黏膜组织中ING2基因表达水平。结果ING2在正常食管黏膜中微弱表达，而在食管鳞癌组织中显著高表达；癌组织中ING2蛋白相对表达量（10．250±3．792）明显高于正常食管黏膜组织（1．368±0．684），差异有统计学意义（P=0．001）。ING2mRNA表达水平显著高于正常食管组织，差异有统计学意义（t=37．310，P=0．001）。结论ING2在食管鳞癌中高表达，且与临床病理因素及预后明显相关。

Objective To investigate the expression of growth inhibitory factor 2 （ING2） in e- sophageal squamous cell carcinoma （ESCC）, and explore its correlation with lymphatic metastasis and prognosis. Methods The expression of ING2 in 63 cases of ESCC tissues and 63 cases of normal tissues was detected by immunohistochemistry. Real - time quantitative reverse transcriptase - polymerase chain reaction （ RT - qPCR） was used to examine the expression of ING2 mRNA in fresh tumoral and normal tis- sues. Results The ING2 positive staining was predominantly localized in both nucleus and cytoplasm of cancer cells, but weakly in nucleus of normal epithelial cells only. The expression of ING2 protein and mRNA was significantly higher in ESCC tissues than in normal tissues （P = 0. 001 ）. Moreover, the expres- sion level of ING2 protein in ESCC tissues was significantly correlated with grade （P = 0. 010） and lymph node metastasis （P =0. 001 ）. Cox regression analysis revealed that ING2 expression level （P = 0. 042 ） and lymph node metastasis （ P = 0. 000） were independent prognostic factors of ESCC. Conclusion ING2 is overexpressed in ESCC tissues. High expression of ING2 correlates with pathological grades, lymphatic metastasis and unfavorable survival in ESCC.
