有氧运动训练对ApoE^(—/—)小鼠肝脏铁调素蛋白和血清IL-6、MCP-1含量的影响 被引量:2

Effects of Aerobic Exercise on the Expression of Hepatic Hepcidin,and the Level of Serum IL-6 and MCP-1 in ApoE^(—/—) Mice

摘要 目的:观察有氧运动训练对载脂蛋白E基因缺陷(ApoE^(—/—))小鼠肝脏铁调素(Hepcidin)蛋白表达及血清IL-6、MCP-1水平的影响,探讨有氧运动防治ApoE^(—/—)小鼠动脉粥样硬化(AS)的作用。方法:20只8周龄雄性ApoE^(—/—)小鼠随机分为对照组(C组,10只)和运动组(E组,10只),对照组小鼠不运动,运动组小鼠进行14周递增负荷跑台训练,每周5次,即小鼠由第1~5周10 m/min,30 min,逐渐递增至13 m/min,60 min,跑台坡度为0°。14周运动干预结束后取材,对小鼠主动脉进行包埋并进行冰冻切片,然后经苏木精-伊红(HE)和油红O(ORO)染色,观察主动脉的病理变化,采用Western blot方法检测肝脏Hep-cidin蛋白表达水平,同时采用双抗夹心酶联免疫(ABC-ELISA)方法检测血清Hepcidin、白细胞介素-6(IL-6)及单核细胞趋化蛋白-1(MCP-1)含量。结果:对照组小鼠主动脉血管壁纤维板断裂,可见大量泡沫细胞浸润和纤维斑块,运动组小鼠较对照组主动脉病变减轻;运动组小鼠Hepcidin蛋白表达及血清水平显著低于对照组(P<0.01);运动组小鼠血清IL-6、MCP-1水平显著低于对照组(P<0.01)。结论:长期有氧运动训练降低了ApoE^(—/—)小鼠肝脏Hepcidin蛋白表达和血清水平,同时降低了小鼠血清中炎症因子IL-6、MCP-1的水平,从而发挥了防治ApoE^(—/—)小鼠动脉粥样硬化的作用。 Objective To observe the preventive effects of the aerobic exercise on the expression of hepatic hepcidin,and the level of serum IL-6 and MCP-1 in ApoE-/- rats with atherosclerosis (AS) and to explore the mechanism. Methods Twenty 8-week male ApoE-/- mice were randomly divided in- to a control group and an exercise group,each of 10. The AS model was established in both groups through feeding high-fat diet. The control group was not given exercise intervention,while the exercise group underwent 14-week progressive intensity treadmill training,5 times a week,starting at 10 m/min for 30 min for the first 5 weeks and increasing to 13 m/min for 60 min at the end. At the end of the experiment,aortic samples were fixed and treated using the hematoxylin-eosin staining and Oil Red O. Then the pathological changes of the aorta were analyzed. The hepcidin expression of liver and se- rum hepcidin was assessed using Western blotting, and the level of the, interleukelin-6 (IL-6) and monocyte chemotactic protein-1 (MCP-1) were detected using ABC-ELISA. Results The pathological slice showed that the vascular fiber plate and internal membrane of the aorta in the exercise group were better than that of the control group, with less ruptured vascular fibreboards, foam cell infiltration and fiber plaques. The hepcidin expression of liver and the level of serum hepcidin, the level of the se- rum IL-6 and MCP-1 in the exercise group were significantly lower than those of the control group (P〈0.01 and P〈0.01). Conclusion The long-term aerobic exercise can prevent AS for ApoE-/ mice through lowering the expression of the hepatic hepcidin and serum hepcidin, as well as the level of se- rum IL-6 and MCP-1.
