花生芽组织块悬浮培养产辅酶Q_(10)的研究 被引量:2

摘要 【目的】建立花生芽组织块培养合成辅酶Q_(10)(CoQ_(10))方法,为高效生产CoQ_(10)提供参考。【方法】以花生芽组织块为试验材料,考察培养温度、培养时间、蔗糖质量浓度、初始pH值对CoQ_(10)合成的影响。在单因素试验基础上,通过L9(34)正交试验优化培养条件。【结果】花生芽经悬浮培养后,其产CoQ_(10)的最优条件为:培养温度24℃,时间20h,蔗糖质量浓度30g/L,初始pH值6.0,在此条件下CoQ_(10)产量最高,为182.147μg/g,比未经悬浮培养处理花生芽产CoQ_(10)含量提高了72.4%。【结论】建立了花生芽组织块悬浮培养产CoQ_(10)的最佳工艺,且这种工艺操作简便、周期短、成本低。 【Objective】The method for synthesis of coenzyme Q_(10) from peanut tissue pieces in suspension culture was proposed.【Method】The effects of temperature,cultural time,sucrose concentration and pH value on synthesis of coenzyme Q_(10) from tissue pieces of peanut buds were investigated to select optimal culture conditions using orthogonal L9(34)test.【Result】The production of coenzyme Q_(10) in peanut tissue pieces was up to 182.147μg/g under the obtained optimal culture conditions of temperature 24℃,cultural time 20 h,sucrose concentration 30g/L,and pH value 6.0.It was 72.4% higher than the production without suspension culture.【Conclusion】The established optimal conditions for producing coenzyme Q_(10) from peanut tissue pieces in suspension culture was featured by short production cycle,simple operating procedure,and low cost.
