艾灸“肺俞、膏肓俞”对BLMA5所致肺纤维化大鼠肺上皮细胞E-cad基因表达的实验研究 被引量:8

摘要 目的:探究艾灸"肺俞、膏肓俞"抑制肺纤维化进程的作用机制。方法 :将81只8周龄SPF级大鼠随机分9组:空白对照组;模型组(7 d组、11 d组、23 d组、35 d组);艾灸组(11 d组、23 d组、35 d组);阳性对照(泼尼松)组,每组9只。采用气管内一次性注入博来霉素复制肺纤维化模型。各组造模均于第二天进行治疗及干预。艾灸组交替艾灸双侧肺俞和膏育俞,3壮/穴,5 mg/壮,1次/d,10次/疗程。泼尼松组以泼尼松灌胃。空白组和所有模型组与艾灸组采取同样的抓取和固定,不给予任何治疗。共治疗3个疗程,疗程间休息2 d。各组在设定的时间窗口处死动物,采用定量PCR法检测E-cad基因的表达水平。结果:艾灸治疗组E-cad基因表达量比同期模型组高,尤以第35 d明显。艾灸35 d组(0.32±0.07)明显高于模型35 d组(0.10±0.04)(P<0.01)。艾灸组和泼尼松组无明显差异,统计学无意义(P>0.05)。结论:艾灸"肺俞、膏肓俞"可抑制肺纤维化的进程,其内在机制可能与升高肺上皮细胞E-cad基因表达有关。 Objective: To investigate the mechanism of moxibustion at Feishu( BL13) and Gaohuang( BL43) on bleomycin-induced pulmonary fibrosis. Methods: Eighty-one healthy male SD rates were randomly divided into nine groups: blank control group,model group( 7 d group,11 d group,23 d group,35 d group),moxibustion group( 11 d group,23 d group,35 d group),positive control group( prednisone group)( n = 9). The exprimental pulmonary fibrosis rat model was induced by a single injection of the bleomycin via trachea. Treatment and intervention were carried out on second day. Moxibustion group was alternatively given moxibustion at bilateral Feishu( BL13) and Gaohuang( BL43),3 Zhuang/point,5 mg/Zhuang,once a day,10 times a course of treatment. Prednisone group was treated with prednisone orally. But blank control group did not give any treatment. There was a total of 3 courses and 2 days rest between the course of treatment. The animals were sacrificed at the time window and the expression level of E-cad gene was detected by quantitative PCR method. Results: The expression of E-cad in the treatment group was higher than that in the model group in the same period,especially on the thirty-fifth day. Moxibustion 35 d group( 0. 32 ± 0. 07)was significantly higher than that of model 35 d group( 0. 10 ± 0. 04)( P〈0. 01). Moxibustion group and prednisone group had no significant difference( P〈0. 05). Conclusion: Moxibustion at Feishu( BL13) and Gaohuang( BL43) can inhibit the progression of pulmonary fibrosis and the underlying mechanism may be related to the increased expression of E-cad gene in lung epithelial cells.
