摘要
建立人舌鳞癌细胞体外癌变模型,采用激光共焦显微拉曼光谱仪对其进行检测,从分子层面对其进行分析.通过培养CAL-27细胞,然后注射在裸鼠皮下成瘤,采用激光共焦显微拉曼光谱仪和苏木精-伊红(HE)染色法观察其癌变的情况.结果表明,正常皮肤组织的切片清晰可见,其细胞层次清晰、整齐,但是肿瘤组织发现上皮异常增生,细胞多形性,结构层次非常不清楚;采用共焦显微拉曼光谱技术,结合主成分分析法和线性判别技术对健康裸鼠和人舌鳞癌细胞体外癌变组织进行分类,其得到诊断灵敏度为98.3%,特异性为95.0%,诊断正确率为96.7%.该方法可以很好地对该动物模型进行在体的检测.
The aim of this paper is to establish a carcinogenesis model of human squamous tongue cancer cell in vitro.The model is tested by laser confocal micro Raman spectrometer and is analyzed from the molecular level.The CAL-27 cells are cultivated, and injected subcutaneously in nude mice into a tumor, then using laser confocal micro Raman spectrometer and Hematoxylin-eosin (HE) staining, the canceration is obtained.From the normal skin tissue slice, it can be seen that the cellular level is clear and neat.But it can be found that the tumor epithelial dysplasia is cellular pleomorphism and the structure level is not clear.After laser confocal micro Raman spectrometer observing, the principal components analysis (PCA) combination with linear discrimination analysis (LDA) are used to classify the health nude mice and human tongue squamous cancer cells in vitro tumor tissue.It can be obtained the diagnostic sensitivity of 98.3%, specificity of 95.0% and accuracies of 96.7%.It is shown that this method is very good for detecting the animal model in vivo.
