银耳发酵液中银耳多糖的醇析条件优化

摘要 本课题以银耳发酵液为样品,以银耳粗多糖含量为评价指标,采用醇析法沉淀银耳多糖、蒽酮比色法测定银耳多糖含量对银耳发酵液中银耳多糖的醇析条件进行研究,为银耳多糖的进一步开发和利用提供参考。探讨了醇析温度、醇析时间、添加乙醇浓度对银耳发酵液中银耳多糖析出的影响,通过单因素实验确定了醇析的最佳条件。结果表明:银耳发酵液中银耳多糖适宜的醇析条件为醇析温度常温,醇析时间28 h,添加乙醇浓度90%。 Taking Tremella fuciformis fermentation broth as sample,the content of crude polysaccharide as the evaluation index,ethanol precipitation method for precipitating polysaccharide and anthrone colorimetric method for measuring the content of polysaccharide were adopted to study the condition required for ethanol precipitation of polysaccharide of Tremella fuciformis fermentation broth,which helped providing a reference for the further development and utilization of Tremella fuciformis polysaccharide. The effects of adding ethanol concentration,ethanol precipitation time and ethanol precipitation temperature on the precipitation of polysaccharide of Tremella fuciformis fermentation broth were investigated. The optimal conditions were determined by single factor experiments. The consequence demonstrated that the optimum condition required ethanol precipitation temperature of normal temperature,ethanol precipitation time of 28 h and adding ethanol concentration of 90%.
