植物乳杆菌YM-4-3发酵合成γ-氨基丁酸的条件优化 被引量:3

Optimization of Fermentation Conditions for γ-Aminobutyric Acid(GABA) Production by Lactobacillus plantarum YM-4-3

摘要 通过单因素分析和正交试验,对Lactobacillus plantarum YM-4-3菌株发酵合成γ-氨基丁酸(gamma-aminobutyric acid,GABA)的培养基配方及各项发酵条件进行优化.优化结果表明,L.plantarum YM-4-3菌株发酵生产GABA的最适发酵条件为:接种量1%,培养基初始pH 6.0,35℃厌氧静置发酵96 h;最佳培养基配方为:Lab-lemco'powder,4 g/L;蛋白胨,15 g/L;酵母浸膏,6 g/L;Tween-80,1 mL/L;磷酸氢二钾,2.0 g/L;蔗糖,20.0 g/L;四水硫酸锰,0.05 g/L;柠檬酸三铵,2.0 g/L;七水硫酸镁,0.2 g/L;乙酸钠,5.0 g/L;谷氨酸钠(monosodium glutamate,MSG)30 g/L(即3%,w/v).在最佳发酵工艺条件下,L.plantarum YM-4-3菌株发酵液中GABA含量可高达15.09 mmol/L. The single factor analysis and orthogonal design experiment are employed to determine the optimal cultivation conditions and optimal medium components for production of GABA by Lactobacillus plantarum YM-4-3 strain.The content of GABA is up to 15.09 mmol/L after the optimization.The optimum cultivation conditions are as follows:inoculation volume is 1%,the initiation pH is 6.0,and static anaerobic fermentation lasts for96 h at 35℃.The optimum culture medium are listed as:4 g/L Lab-lemco' powder,15g/L Peptone,6 g/L Yeast extract,1mL/L Tween-80,2.0 g/L Di-potassium hydrogen phosphate,20.0 g/L Sucrose,0.05 g/L Manganese sulphate·4H2O,2.0 g/L Tri-ammonium cirtrate,0.2 g/L Magnesium sulphate·7H2O,5.0 g/L Sodium acetate,and 30g/L monosodium glutamate(MSG)(3%,w/v).
