BMP诱导活体内肌腱组织成骨化的研究 被引量:2

Bone morphogenetic protein-induced ossification of tendinous tissue in vivo

摘要 [目的]探讨活体内BMP诱导肌腱组织成骨化的效果。[方法]30只新西兰兔建立活体内肌腱组织诱导成骨模型,一侧为rh BMP-2处理、对侧为空白对照。术后4、8及12周时采用X线片检测肌腱组织钙化情况,术后12周时对肌腱组织进行大体解剖、组织形态及免疫组化检测分析。[结果]术后4、8及12周时X线片检测发现BMP处理侧各时间点上肌腱组织内可见钙化区域,成骨化范围随时间增加逐渐增大(5.51 mm2、9.44 mm2及20.26mm2,P<0.05),钙化区域IOD值亦随时间增加而增高(1 178.56、2 135.26及4 726.72,P<0.05)。大体解剖可见BMP处理组肌腱组织增粗、质地变硬,H.E及Masson染色检测见肌腱纤维内细胞浸润,局部区域骨样组织形成,并有osteonectin阳性表达细胞分布。此外,肌腱纤维及骨化区域内均可见软骨样细胞形成。[结论]BMP能够诱导活体内肌腱组织发生成骨化,可能与软骨内成骨途径有关。 [Objective] To investigate the ossifieation of tendinous tissue induced by bone morphogenetic protein (BMP) in vivo. [Method ] Thirty New Zealand rabbits were involved in this study. For eaeh animal the tendon of semitendinosus in one limb was treated by rhBMP-2, while the contralateral tendon was treated by PBS. At 4, 8 and 12 weeks after the treatment, the ossification of tendinous tissue was evaluated by X-rays. Additionally, ealcified tissue were investigated by the gross anatomy, histomorphology and immunohistochemistry at 12 weeks. [Resultl The ossification of tendinous tissue treated with BMP was found by X-rays at 4, 8, and 12 weeks. The area of calcified tissue gradually increased over time (5.51 mm^2. 9.44 mm^2 and 20.26 mm^2P〈0.05) , and the IOD of ossifieated area also increased correspondingly ( 1 178.56, 2 135.26 and 4 726.72, P〈0.05) . The ossificated tendon had a larger diameter and harder texture in gross observation. In lerm of histomorphology, the H.E and Masson staining showed cellular infihration around the tendon fibers and osteoid tissue with osteonecfin-posilive cells in some area. Furthermore, ehondroid cells were deteeted among tendon fibers. [Conclusion] The BMP can imtm'e ossification of tendinous tissue in vivo, which is likely to be related to endochondral ossification.
