摘要
目的:研究麻黄细辛附子汤对变应性鼻炎(allergic rhinitis,AR)大鼠INF-γ与IL-13的影响,揭示麻黄细辛附子汤治疗AR的作用机制。方法:将40只wistar大鼠按体质量采用随机区组法分成5组,即正常对照组、模型对照组、扑尔敏治疗组、麻黄细辛附子汤低剂量组、麻黄细辛附子汤高剂量组,每组8只,造模分为2个阶段,即基础致敏与鼻内激发。基础致敏:实验组使用5%OVA混悬液致敏,腹腔注射,日1次,共7次;对照组以等量生理盐水替代5%OVA混悬液,方法和步骤同实验组。鼻内激发:实验组从第15天起,以5%卵清蛋白(ovalbumin,OVA)混悬液100μL双侧滴鼻,每侧50μL,日1次,共7次;对照组以等量生理盐水替代5%OVA混悬液,方法和步骤同实验组。麻黄细辛附子汤低剂量组:使用麻黄细辛附子汤2 m L·次^(-1)(每毫升含生药0.2 g),1日2次灌胃,共21 d。麻黄细辛附子汤高剂量组:以麻黄细辛附子汤2 m L·次^(-1)(每毫升含生药0.4 g),一日2次灌胃,共21 d。模型组及正常对照组分别使用生理盐水2 m L·次^(-1),日2次灌胃,共21 d。扑尔敏治疗组:0.01%扑尔敏溶液2 m L·次^(-1),日2次灌胃,共21 d。检测麻黄细辛附子汤治疗后血浆INF-γ与IL-13的含量。结果:模型组INF-γ含量(17.4±2.7)μg·L-1显著降低,IL-13(50.3±2.6)μg·L-1显著升高,治疗后麻黄细辛附子汤低剂量组和麻黄细辛附子汤高剂量组INF-γ含量(22.6±3.0;20.4±2.5)μg·L-1含量升高,IL-13(35.0±3.0;34.2±3.3)μg·L-1含量降低。模型组与正常对照组比较有统计学意义(P<0.01)。麻黄细辛附子汤治疗组、扑尔敏治疗组与模型组比较有统计学意义(P<0.01)。AR大鼠INF-γ含量较低,而IL-13含量较高,与正常对照组比较有统计学意义(P<0.01)。结论:麻黄细辛附子汤可以调节免疫失衡功能,通过使INF-γ含量升高,使IL-13含量降低纠正体内Thl和Th2表达失衡而发挥疗效。
Objective: To study the influence of Ephedra and Aconite and Asarum Decoction on contents of INF-γ and IL-13 of rats with allergic rhinitis, and reveal its mechanism in the treatment of allergic rhinitis (AR) . Methods: 40 wistar rats were divided into 5 groups according to weight by using random blocking method: normal control group, model control group, chlorpheniramine group, low-dose group of Ephedra and Aconite and Asarum Decoction, high-dose group of Ephedra and Aconite and Asarum Decoction, with 8 rats in each group. The model-establishing was divided into 2 stages, namely the basic sensitization and intranasal provocation. The basic sensitization: The experimental group was sensitized by 5% OVA suspension, injected intraperitoneally 1 time a day, for 7 times together, while the control group used the same amount of normal saline to replace 5% OVA suspension. The method and procedure were the same as the experimental group. The intranasal provocation: From the fifteenth day, the experimental group was given 100μl of 5% OVA suspension for dripping bilateral nose, each side 50μl, 1 times a day, for 7 times together, while the control group used the same amount of normal saline to replace 5% OVA suspension. The method and procedure were the same as the experimental group. Low -dose group of Ephedra and Aconite and Asarum Decoction used 2 ml of the decoction each time (each milliliter containing crude drug 0. 2 g) for intragastric administration, twice a day, for 21 days together. High -dose group of Ephedra and Aconite and Asarum Decoction used 2 ml of the decoction each time ( each milliliter containing crude drug 0.4 g) for intragastric administration, twice a day, for 21 days together. The model group and normal control group were treated with physiological saline 2 mL each time, twice a day for intragastric administration, for 2 days together. Chlorpheniramine group was treated with 2 mL chlorphenirmnine solution with O. 01% concentration each time, twice a day for intragastric administration, for 2 days together. Then the contents of INF- gamma and IL-13 in plasma were detected after being treated with Ephedra and Aeonite and Asarum Decoction. Results: The model group' s INF-gamma content ( 17. 4 ±2.7) ng·L^-1 decreased significantly, IL-13 (50.3± 2.6) ng ·L^-1 increased significantly. After the treatment, the contents of INF-gamma in low-dose group of Ephedra and Aconite and Asarum Decoction, and high-dose group of Ephedra and Aconite and Asarum Decoction: (22:6± 3.0;20.4 ± 2.5) ng·L^-1 increased, while the contents of IL-13 ( 35 ± 3 ; 34.2 ±3.3 ) ng · L^-1 decrased. Compared with the normal Control group, the model group was statistically significant (P 〈0.01 ), Ephedra and Aco- nite and Asarum Decoction treatment groups, chlorpheniramine group, and model control group were statistically significant ( P 〈 0. 01 ). The content of INF- gamma in AR rats was lower, but the content of IL-13 was higher, which was statistically significant compared with the normal control group (P 〈0.01 ). Conclusion: Ephedra and Aconite and Asarum Decoction can modulate the imbalance of immune function, correct the imbalance of Thl and Th2 expression in the body by increasing the content of INF- gamma, and decreasing the content of IL-13.
