摘要 目的:采用HPLC法对广西荔浦地区的牛耳枫药材进行指纹图谱研究。方法:利用Agilent HP 1260,测定同一地区的12批牛耳枫药材。Waters XBridge^(TM)C_(18)色谱柱(250 mm×4.6 mm,5μm),乙腈(A)-0.1%磷酸溶液(B)为流动相进行梯度洗脱,流速1.0 mL/min,柱温35℃,检测波长254 nm。结果:建立了广西荔浦地区的牛耳枫药材的HPLC指纹图谱,标定了11个共有峰,指认了2个峰,各指纹图谱的相似度均达到0.85以上;利用系统聚类分析,将同一地区的牛耳枫分为3类;利用主成分分析,确定3个主成分,累计贡献率为81.980%;以两个主成分作二维散点图,将不同批次的牛耳枫分为3大类。结论:运用这3种统计分析方法均能够较好的对指纹图谱进行识别,为牛耳枫的鉴别、质量控制等提供了新的科学依据。 Objective : To establish the high performance liquid chromatography fingerprint of Daphniphyllum calycinum from Lipu County of Guangxi. Methods:Agilent HP 1260 was applied to analyze twelve batches of Daphniphyllum calycinum. Waters XBridgeTM C185 column was performed(250 mm × 4. 6 mm,5 μm)with acetonitrile(A)-0. 1% phosphoric acid (B)in agradient mode at a flow rate of 1.0 mL/min, the column temperature was 35 ℃. The detection wave length was 254 nm. Results : The HPLC fingerprint of Daphniphyl- lum calycinum were established with calibrated eleven common peaks, and two of them were identified. The similarity was greater than 0. 85. The cluster analysis was adopted to divide twelve batches samples into three categories. Three principal components were observed by principal component analysis and the value of three principal components contributed 81. 980%. The two main components of them were employed to plot a two-dimensional scatter map and twelve batches samples were divided into three categories. Conclusion:The three employed methods can reflect the similarity of the fingerprint accurately. Those recognition modes described can provide an identi- fication method for fingerprint, and provide the foundation of quality control.
