基于JNK信号通路的蜂毒穴位注射抑制Lewis小鼠肺癌肿瘤血管生成 被引量:3

Injecting for Inhibiting the Formation of Tumor Vessel of Lung Cancer of Lewis Mice Based on the Point Injection of Bee Venom of JNK Signal Pathway

摘要 目的:探讨蜂毒穴位注射抑制肿瘤血管生成作用及其相关机制。方法:建立C57BL/6J Lewis肺癌小鼠模型;隔日测量肿瘤最大径与最小径,计算肿瘤体积,绘制生长曲线;第24天麻醉取材称量瘤质量;解剖显微镜下计数肺上转移结节;采用免疫组化法检测CD34计算肿瘤微血管密度;采用Western Blotting法观察药物对JNK信号通路相关蛋白的影响;采用qPCR法观察HIF-1α和VEGF mRNA的表达。结果:与模型组比较,各治疗组肿瘤体积、瘤质量、肺上转移结节、肿瘤微血管密度、p-JNK、p-c-Jun、HIF-1α、VEGF蛋白及HIF-1α、VEGF mRNA表达均显著降低,且蜂毒穴位注射组的疗效优于穴位组及蜂毒非穴位组。结论:蜂毒穴位注射能抑制肿瘤血管生成,其作用机制与调控JNK信号传导有关。 Objective:To investigate the effect of point injection with bee venom, and to study the relevant mechanism. Methods: C57BL/6J Lewis lung cancer mice models were constructed;and measured the maximum and minimum diameter of tumors on alternate day, the volume of tumors were calculated, and the growth curve was drawed. In the 24th day, anaesthetized the draw materials and weighed the tumor, counted the pulmonary metastatic nodules by the dissecting microscope;while CD34 was detected by calculating the microvessel density in immunohistochemistry. The drug effect on relative protein of JNK signal passage was detected by Western blot- ting;the expression of HIF-1α and VEGF mRNA was detected by qPCR method. Results:Compared with the model group, the tumor vol- ume,the tumor weight and the lung metastatic nodules, the tumor microvessel density and the expression of the p-JNK, p-c-Jun, HIF-1α,VEGF proteins and HIF-1α and VEGF mRNA were decreased significantly and with effect on the point injection with bee renom group were better than the point group and the non-point injection group. Conclusion:Point injection with bee venom can inhabit angio- genesis, and the mechanism of effect may be relate to the regulation of JNK signal pathway.
