摘要
目的探究A型流感病毒复制过程中特属于人类宿主的专一性调控因子。方法利用LNA-1290反证hsamiR-1290对vimentin的调控,证明hsa-miR-1290对VIM抑制的专一性。结果人类特异的hsa-miR-1290在A549细胞经A/WSN/33(H1N1)感染后,表现量在单一与多重性感染的第2 h与24 h后,分别有1.9倍与5.5倍的增加。hsa-miR-1290的表现导致A/WSN/33(H1N1)的病毒蛋白质与产量皆有显著的增加。Vimentin蛋白质为hsamiR-1290的标记基因,其mRNA与蛋白质的表现因hsa-miR-1290的表现而降低至30%与40%。抑制vimentin可以促进A/WSN/33(H1N1)的病毒蛋白质与产量显著的增加。结论 hsa-miR-1290可以通过抑制VIM帮助A/WSN/33(H1N1)在A549人类细胞中的复制。
Objective To explore the specific regulatory factors of human host in the replication process of influenza A virus.Methods The regulation of vimentin by hsa-miR-1290 was disproved by LNA-1290,which demonstrated the specificity of hsa-miR-1290 on VIM inhibition. Results After the specific hsa-miR-1290 gene of human infected with A/WSN/33( H1N1),the expression of hsa-miR-1290 was 1. 9 and 5. 5 times higher than that of single and multiple infection in 2 h and24 h. The expression of hsa-miR-1290 resulted in a significant increase in the viral protein and yield of A/WSN/33( H1N1). Vimentin protein was a marker gene for hsa-miR-1290,whose mRNA and protein are reduced by 30% and 40%due to hsa-miR-1290. Suppression of vimentin can contribute to a significant increase in A/WSN/33( H1N1) viral protein production. Conclusion hsa-miR-1290 can help A/WSN/33( H1N1) replication in A549 human cells by inhibiting VIM.
