摘要
目的对婴儿配方奶粉生产加工过程中的原料和环境中分离的阪崎克罗诺杆菌和铜绿假单胞菌开展PFGE分子分型研究,分析污染途径。方法在一个批次婴儿配方奶粉生产加工过程中的原料和环境中分别检出了阪崎克罗诺杆菌和铜绿假单胞菌。用限制性内切酶XbaⅠ酶切阪崎克罗诺杆菌,用SpeⅠ酶切铜绿假单胞菌后进行PFGE电泳,并用Bio Numerics软件进行聚类分析。结果 4株铜绿假单胞菌的基因指纹图谱完全一致,为同一来源菌株;4株阪崎克罗诺杆菌分为2个基因型。结论原料、环境、设备和车间工人之间都存在交叉污染,原料是污染的主要源头;环境和设备的CIP清洗死角是污染的藏匿地;车间工人很可能是污染扩散的主要媒介。
Objective To study on PFGE molecular typing of Enterobacter sakazakii Craunot bacillus and Pseudomonas aeruginosa isolated from infant formula in the process of production and processing of raw materials and the environment,and then pollution pathways were analyzed. Methods Enterobacter sakazakii Craunot bacillus and Pseudomonas aeruginosa were detected from the same batch of infant formula in the process of production and processing of raw materials and the environment. Enterobacter Sakazakii was digested with enzyme XbaⅠand Pseudomonas aeruginosa was digested with enzyme SpeⅠ. Then PFGE electrophoresis was performed and Bio Numerics software was used for cluster analysis. Results Gene fingerprintings of 4 strains of Pseudomonas aeruginosa were completely consistent,and they were of the same origin; 4 strains of Enterobacter sakazakii Craunot were divided into two genotypes. Conclusion There are cross contamination between raw materials,environment,equipment and workshop workers. Raw materials are the main source of contamination; environmental and equipment CIP cleaning dead corner is a contaminated hiding place; workshop workers are likely to be the media of pollution diffusion.
