电针内关穴、天枢穴对肠易激综合征模型大鼠影响的对比研究 被引量:8

摘要 目的:通过电针肠易激综合征(IBS)模型大鼠内关穴、天枢穴后,观察大鼠肠道疼痛敏感反应、情绪反应的变化,结合肠道辣椒素受体(TRPV1)阳性表达的变化,探讨不同穴位对IBS不同症状及肠道机制调节的效应差异,从而进一步阐释经穴效应特异性的部分效应机制。方法:将Wistar新生鼠在造模前分为空白对照组(以下简称为空白组)和模型组,除空白组外,模型组在Wistar新生鼠出生第2天后分别予以2周的醋酸灌肠及母子分离,并在其6周龄时予以肠道球囊扩张刺激3次。造模结束后,将模型组随机分为模型对照组(以下简称模型组)、内关组、天枢组。空白组及模型组大鼠不做任何处理,内关组及天枢组分别予以相应穴位的电针治疗,隔日1次,共5次。采用腹部回撤反射实验和旷场实验来评价各组大鼠内脏敏感性和情绪心理行为;采用免疫组织化学方法检测大鼠结肠中辣椒素受体免疫阳性表达水平。结果:与空白组比较,模型组大鼠出现疼痛反应的潜伏期明显缩短及收缩波个数明显增多、水平运动次数以及垂直运动次数均明显减少、结肠内TRPV1阳性表达水平明显增强(P<0.01);与模型组比较,内关组大鼠的疼痛潜伏期延长、水平运动次数增加、TRPV1阳性表达水平明显减弱(P<0.05;P<0.05;P<0.01),且天枢组大鼠的疼痛潜伏期明显延长、收缩波个数明显减少、结肠内TRPV1阳性表达水平明显减弱(P<0.01);两治疗组比较,天枢组大鼠的疼痛潜伏期较内关组明显延长(P<0.01),差异具有统计学意义。结论:电针内关、天枢穴对IBS的肠道敏感性及情绪心理作用存在效应特异性,而在结肠内辣椒素受体的表达上未表现出差异。 Objective: By observing the IBS model rats' intestine sensitivity of pain reaction,emotional reaction and the expression change of TRPV1,to discuss the different affection to the model rats' corresponding symptoms and analyze part of function mechanism of the specific of acupoints affection through electro-acupuncture on Neiguan( PC6) and Tianshu( ST25). Methods: Wistar rats were randomly divided into blank group( BG),model group( MG),Neiguan( PC6) electro-acupuncture group( NG) and Tianshu( ST25) electro-acupuncture group( SG). Except the BG,the other groups' newborn rats received colonic irritation by acetic acid,were separated from their mother from postnatal day2 to day 15 and given intestinal stimulation by dilated balloon three times at 6 weeks old. Results: Comparing with the BG,the MG shortened the latency of pain,increased the number of contraction wave,decreased the the scores in horizontal motion and vertical motion and increased the expression of TRPV1( P〈0. 01). Comparing with MG,the NG prolonged the latency of pain,increased the scores in horizontal motion and decreased the expression of TRPV1( P〈0. 05,P〈0. 05,P〈0. 01). Comparing with MG,the TG prolonged the latency of pain and decreased the number of contraction wave significantly,increased the scores in horizontal motion and decreased the expression of TRPV1( P〈0. 01). Comparing with NG,the TG prolonged the latency of pain significantly( P〈0. 01). Conclusion: The effect of rats' intestine sensitivity of pain reaction and emotional psychological abnormalities through electro-acupuncture on PC6 and ST25 is different,but that did not reflect the expression of TRPV1.
