红景天甲醇冷浸提取物的体外抗病毒谱及抗病毒机制的研究 被引量:12

摘要 目的:为建立红景天抗病毒谱-效相关评价系统,对红景天抗病毒机制进行初步探究,通过对红景天甲醇冷浸提取物体外抗病毒活性进行筛选,最后选择红景天甲醇冷浸提取物抗柯萨奇病毒B型(COX-B5)活性进行研究病毒机制的初步研究。方法:采用体外抗病毒实验CPE法结合MTT染色法,以半数治疗浓度(EC50)和半数细胞毒性浓度(TC50)并以治疗指数TI值作为评价指标,观察红景天甲醇冷浸提取物对手足口病病毒71型(EV71)、柯萨奇病毒B型(COX-B5)、单纯疱疹病毒Ⅰ型(HSV-1)和呼吸道合胞病毒(RSV)的抑制作用。结果:中药材红景天对于四种病毒感染都有一定程度的抑制效果,其中以肠道病毒的抑制效果最好,红景天甲醇提取物对呼吸道合胞病毒(RSV)的治疗指数为26.35,对单纯疱疹病毒Ⅰ型(HSV-1)的治疗指数为14.03,对柯萨奇病毒B型(COX-B5)的治疗指数为33.82,对手足口病病毒71型(EV71)的治疗指数为27.28。在此基础上,选取红景天甲醇冷浸提取物抗柯萨奇病毒B型(COX-B5)活性做进一步的抗病毒机制研究,结果发现,红景天甲醇冷浸提取物在预防性治疗中,其治疗指数TI值为11.31,在抑制病毒穿入感染中,其治疗指数为19.29,在药物病毒直接作用的灭杀组中,药物的治疗指数为35.26。结论:中药材红景天对于肠道病毒的抑制有明显的效果,但其作用机制较复杂,并非针对病毒吸附感染复制的某一单一环节,作用机制有待进一步研究,红景天作为低毒高效的传统中药材,在抗病毒领域有很大潜力,有待进一步研究开发。 Objective: To establish a rhodiola antiviral spectrum-related evaluation system and carry on the preliminary exploration of rhodiola antiviral mechanism by means of screening methanol soaking extract of rhodiola rosea in vitro antiviral activity and finally choose methanol soaking extract of rhodiola rosea against coxsackie virus B( COX-B5) to study the active virus mechanism. Methods: The vitro antiviral experiment used CPE method combined with MTT staining method,half concentration( EC50),half cell toxicity concentration( TC50) and TI as evaluation indexes. Observe methanol soaking extract of rhodiola rosea for hand,foot and mouth disease virus type 71( EV71),coxsackie virus B( COX-B5),herpes simplex virus type Ⅰ( HSV-1) and inhibition of respiratory syncytial virus( RSV). Results: Traditional Chinese medicine rhodiola rosea has a certain degree of inhibition effect and the best effect was on the intestinal virus.The TI value of respiratory syncytial virus( RSV) index was 26. 35 and that of type Ⅰ herpes simplex virus( HSV-1)in the treatment of index was 14. 03 and that of coxsackie virus B( COX-B5) in the treatment of index was 33. 82. For hand,foot and mouth disease virus type 71( EV71),the TI index was 27. 28. On this basis,methanol soaking extracts against coxsackie virus B( COX-B5) antiviral mechanism research showed that the therapeutic index TI was 11. 31. In inhibiting virus its therapeutic index was 19. 29. In direct action group,the drug treatment index was 35. 26. Conclusion:Chinese herbal medicine rhodiola rosea has obvious effect for intestinal virus inhibition,but its mechanism is complicated. The adsorption mechanism needs further study. As low toxicity and efficiency,rhodiola rosea has great potential in the field of antiviral effect and needs further research and development.
