摘要
实验模拟机体消化道环境,采用单因子实验设计,在体外用酶制剂水解DDGS,测定反应后还原糖的生成量,旨在从市面上A、B、C三家企业的木聚糖酶和纤维素酶中筛选出对DDGS水解效果最好的酶制剂,确定比较合理的酶制剂添加水平,并探索纤维素酶和木聚糖酶的配合效应,为非淀粉多糖酶在肉鸡日粮中的运用提供理论基础.结果表明:A的木聚糖酶和纤维素酶优于其他企业的酶;每5克DDGS木聚糖酶最适添加量为181u,纤维素酶最适添加量为628.5u,而且纤维素酶对DDGS的水解作用要比木聚糖酶强;在纤维素酶和木聚糖酶配合水解时,企业A的两种酶的配合效果要优于企业B,在确定木聚糖酶添加量150u后,与不同水平的纤维素酶配合,根据回归方程计算得出复合情况下A的纤维素酶最适添加量为509u.
This experiment simulated conditions of the body's digestive tract and used the enzymes to hydrolyze the DDGS in vitro and determine reducing sugar, which was designed to choose the enzymes that hydrolysis of DDGS works best from xylanases and cellu- loses of A,B and C companies, to determine a reasonable level of enzyme adding in diet, to explore the coordination effect of cellulase and xylanase, to provide the theoretical basis of non-starch polysaccharide enzymes applying in broile~ diet.The results showed that xylanase and cellulase enzyme of A company is better than that of other enterprises. The optimal dosage of xylanase and cellulase is 181U and 628.5U every 5 grams of DDGS. The effect of hydrolysis DDGS of cellulose is stronger than xylanase. In coordination effect of cellulase and xylanase, the effect of enzymes of A company is better than that of B company. When xylanase amount as 150U , then cooperated with different levels of cellnlase, we can get the optimal dosage of cellulase of A company was 509U according to the regression equation.
