中华结缕草对潮霉素的敏感性研究 被引量:1

摘要 研究了中华结缕草3种不同外植体对不同浓度潮霉素溶液的敏感性。结果表明:中华结缕草不同外植体对潮霉素的敏感性存在差异,其中种子经50 mg/L潮霉素处理3周后,发芽率极显著减少,只有6%;愈伤组织经20 mg/L潮霉素处理3周后褐化率达到62%;离体叶片经50 mg/L潮霉素处理7 d后,黄化面积率超过85%。故中华结缕草的种子、愈伤组织、离体叶片在遗传转化中适宜的潮霉素筛选浓度分别为50、20、50 mg/L。 The sensitivities of 3 types of explants of Zoysia sinensis to different concentrations of hygromycin solution were studied. The results showed that the sensitivities of different explants of Z. sinensis to hygromycin were different. Three weeks after the seeds were treated with 50 mg/L hygromycin,their germination rate was very significantly reduced to only 6%. Three weeks after the calluses were treated with 20 mg/L hygromycin,their browning rate reached 62%. Seven days after the isolated leaves were treated with 50 mg/L hygromycin,their yellowing area rate exceeded 85%. Therefore,the optimum concentration of hygromycin for the screening of seed,callus and isolated leaf of Z. sinensis in genetic transformation was 50,20 and 50 mg/L,respectively.
