摘要 为了提高化学法水解猪毛提取角蛋白效率,探讨了采用酶辅助碱解法提取猪毛中角蛋白工艺,并对提取工艺进行了响应面法优化。通过比较盐酸和氢氧化钠对猪毛的水解效果,发现氢氧化钠更有利于角蛋白的提取。通过比较酶辅助氢氧化钠水解猪毛,发现酶辅助碱解猪毛显著(p<0.05)提高了角蛋白的提取效率。采用响应面法对酶辅助碱解工艺进行优化,得出碱浓度和碱解时间的共同作用对角蛋白的提取有显著影响(p<0.05)。拟合显著性及验证结果显示,所建立的拟合模型具有可靠性。根据拟合模型,对酶辅助碱解工艺进行优化,得出碱浓度0.4 mol/L,碱解时间3 h时,酶辅助碱解效果最好。 To improve the efficiency of alkaline hydrolysis to extract keratin from pig hair, effect of enzyme assisted alkaline hydrolysis on extraction ratio of keratin is investigated,and the extraction process is optimized with response surface method.By comparing effect of acid and alkaline hydrolysis on extraction ratio of keratin,it is found that sodium hydroxide is more efficiency for hydrolysis of pig hair. By comparing enzyme assisted sodium hydroxide to hydrolyze pig hair,it is found that enzyme assisted alkali solution significantly(p〈0.05) improved the hydrolysis efficiency. The results of response surface optimization indicate that the combined effect of alkali concentration and alkaline hydrolysis time had a significant effect(p〈0.05)on the hydrolysis of pig hair. The ANOVA of fitting results show that the obtained fitting model is reliable to be used to predict the optimal process of enzyme assisted alkaline hydrolysis of pig hair. According to the fitting model,the enzyme assisted alkali hydrolysis process is optimized,the results show that the extraction ratio of keratin is the highest when the alkali concentration is 0.4 mol/L and the alkaline hydrolysis time is 3 h.
