雷公藤内酯醇对黑色素瘤细胞增殖、凋亡、迁移和侵袭的影响 被引量:2

摘要 目的探讨雷公藤内酯醇(TPL)对体外培养黑色素瘤细胞SK-Mel-28增殖、凋亡、迁移和侵袭的影响。方法以0、20、40、80ng/m L的TPL作用于SK-Mel-28细胞,采用MTT法检测TPL对细胞增殖作用;流式细胞术测定细胞凋亡率;Western blot用于检测细胞凋亡蛋白的表达;Transwell实验用于检测细胞迁移和侵袭。结果随着TPL浓度和作用时间的增加,细胞增殖率逐渐减低(P<0.05),而细胞凋亡率逐渐增加(P<0.05)。Western blot结果显示不同浓度的TPL作用SK-Mel-28细胞后,caspase-3和Bax蛋白表达量显著增加,Bcl-2蛋白的表达量显著减低。TPL处理后,转至Transwell膜下表面的细胞数明显少于对照组。结论 TPL可抑制黑色素瘤细胞增殖、迁移和侵袭,并促进其凋亡,调节凋亡蛋白与促凋亡蛋白的表达,从而在黑色素瘤中发挥抗肿瘤作用。 Objective To explore the effects of triptolide( TPL) on the proliferation,apoptosis,migration and invasion of malignant melanoma cells SK-Mel-28. Methods SK-Mel-28 cells were treated with different levels of TPL( 0ng/m L,20ng/m L,40ng/m L,and 80ng/m L). Then MTT assay was used for detection of cell proliferation ability. Flow cytometry was performed to analyze the apoptosis rate of SK-Mel-28 cells. The expression levels of caspase-3,Bcl-2 and Bax were measured by western blot. Transwell assay was performed to detect the migration and invasion of SK-Mel-28 cells. Results The proliferation rate of SK-Mel-28 cells decreased significantly,while apoptosis rate significantly increased following the rising of dose and affecting time of TPL( P 0. 05). Western blot results showed that the expression levels of caspase-3 and Bax increased,while the expression level of Bcl-2 decreased with the increase of TPL concentrations. After TPL treatment,the number of migrated and invasive cells decreased significantly. Conclusion TPL could inhibit proliferation,migration and invasion,and induce apoptosis of melanoma cells. Additionally,it could regulate the expression of apoptosis-associated proteins to play anti-tumor effects on melanoma.
