摘要
目的探讨外周淋巴器官在成年大鼠胫神经永久性横断伤(即改良型备用性神经损伤,modified spared nerve injury,mSNI)后机械性疼痛异常的慢性转化中的作用。方法 30只成年雄性Sprague-Dawley(SD)大鼠随机分为3组,每组10只动物,分别用于脾脏切除、腰部淋巴结切除和右侧腘窝淋巴结-腹股沟淋巴结联合切除实验。每项实验的10只动物再随机分为外周淋巴器官切除手术组(n=5)和假手术组(n=5)。所有动物根据实验分组进行相应的外周淋巴器官切除的手术或者假手术,术后1周进行右侧mSNI手术。所有动物于外周淋巴器官切除术前、右侧mSNI手术前、mSNI手术后7d和mSNI手术后14d进行von Frey纤维试验,测试mSNI手术同侧和对侧大鼠后爪足底面腓肠神经和隐神经皮肤区域特异性的疼痛反应。结果脾脏和腰部淋巴结切除明显缓解mSNI后7d和(或)14d的大鼠手术侧后爪足底面腓肠神经和隐神经皮肤区域的机械性疼痛异常。然而,右侧胭窝淋巴结-腹股沟淋巴结联合切除对mSNI后7d和14d的大鼠手术侧后爪足底面腓肠神经和隐神经皮肤区域的机碱性疼痛异常没有显著影响。结论脾脏和腰部淋巴结参与成年大鼠mSNI后机械性疼痛异常的慢性转化。
Objective To investigate the role of peripheral lymphoid organs in the chronic transition of mechanical allodynia in adult rats after permanent transection ( modified spared nerve injury, mSNI) of tibial nerves. Methods 30 male adult Sprague-Dawley (SD) rats ( ten weeks old, weighing 250-300g) were randomly divided into three groups, 10 rats each group for splenectomy, lumbar lymphadenectomy and combined right popliteal and inguinal lymphadenectomy, respectively. Each group were then further divided randomly into two groups, of 5 rats in each, one group for peri-lymphadenectomy, and the other group for sham-operation. The rats were subjected to surgical excision of different peripheral lymphoid organs according to the design above, and a week after that to mSNI of the right tibia1 nerves. Von Frey hair test was employed to evaluate the region-specific pain response on the plantar sural and saphenous skin area of both the ipsilateral and eontralateral hindpaws at four different time points: before the excision of peripheral lymphoid or- gans, before mSNI, and 7 and 14 days after mSNI. Results Splenectomy and lumbar lymphadeneetomy significantly alleviated the mechanical allodynia in the sural and saphenous skin area of the ipsilateral hindpaw plantar surface after mSNI 7 and/or 14 days. while combined fight popliteal and inguinal lymphadenectomy had no such effect. Conclusion The spleen and lumbar lymph nodes are involved in the chronic transition of mechanical allodynia after mSNI in adult rats.
