摘要
目的:探讨虾青素对脐静脉内皮细胞(HUVEC)的抗氧化作用。方法:体外培养HUVEC,分为空白组、模型组(H2O22 mmol/L)、虾青素+H2O2组(0.1,1,10μmol/L虾青素预处理48 h后,加2 mmol/L H2O2处理1 h)。用MTT法检测细胞的存活率,DCFH-DA法检测细胞内ROS水平,JC-1法检测线粒体膜电位,Annexin V-FITC流式细胞术和DAPI法检测细胞凋亡,western blot法检测Caspase-3和p53蛋白表达。结果:与空白组相比,H2O2能明显造成HUVEC细胞的凋亡和坏死。虾青素可以降低H2O2引起的细胞死亡,减少活性氧的产生,使线粒体膜电位升高,凋亡率减少,Caspase-3和p53的表达下调。结论:虾青素对H2O2引起的HUVEC细胞死亡具有保护作用,其机制可能与保护线粒体功能有关。
Objective: To explore the antioxidant effects of astaxanthin on human umbilical vein endothelial cells(HUVEC). Methods: HUVEC cells were divided into normal control group, model group(2 mmol/L H2O2), and astaxanthin+H2O2(pretreated with 0.1, 1, 10 μmol/L astaxanthin for 48 h and then cultured with 2 mmol/L H2O2 for 1 h). The cell survival was measured by MTT method, the level of reactive oxygen species was determined by DCFH-DA, the changes of mitochondrial membrane potential and apoptosis ratio were detected by JC-1 method, and Annexin V-FITC and DAPI methods. The expression of Caspase-3 and p53 were measured by western blot. Results: Compared with control group, 2 mmol/L H2O2 obviously caused the necrosis and apoptosis of HUVEC, while astaxanthin could reduce this damage, which was presented by the significantly decreased cell necrosis ratio, increased MMP and down-regulated Caspase-3 and p53 expression. Conclusion: Astaxanthin inhibited oxidative stress induced cell death of H2O2, and its mechanisms may be related to protecting the mitochondrial function.
