摘要
通过评价15种药食同源中药提取物的抗氧化及抗骨质疏松活性,筛选获得具有抗骨质疏松活性的中药。以15种药食同源中药的醇提物和水提物为对象,通过测定提取物的氧自由基吸收能力、还原能力及细胞内抗氧化能力,评价其抗氧化活性。用抗酒石酸酸性磷酸酶(TRAP)染色及活性检测方法评价其抗骨质疏松活性。研究结果表明,3种药食同源中药的5个提取物表现出良好的抗氧化作用,可明显地抑制TRAP活性,包括花椒醇提物、薄荷醇提物及水提物、青果醇提物及水提物,且均以剂量依赖的方式抑制破骨细胞增殖及TRAP活性,当质量浓度为1μg/m L时,薄荷水提物(93%,308%)、青果醇提物(87%,292%)和水提物(83%,297%)对破骨细胞增殖及TRAP活性的抑制效果较好,具有较强的抗骨质疏松活性。本文可为药食同源中药抗骨质疏松保健功能的开发提供理论基础。
Edible traditional Chinese medicine that possessed the anti-osteoporosis activity were obtained by evaluating the antioxidant and anti-osteoporosis activities of the extracts of fifteen edible traditional Chinese medicine in this study. The antioxidant activity of the Et OH and H2O extracts of fifteen edible traditional Chinese medicine was assessed using oxygen radical absorbance capacity, reducing capacity and cellular antioxidant capacity assays. And their anti-osteoporosis activity was further determined by tartrate-resistant acid phosphatase(TRAP) staining and activity assay. These results indicated five extracts of three edible traditional Chinese medicine had favorable antioxidant activity and suppressed TRAP activity, including Et OH extract of Zanthoxylum Bungeanum Maxim., Et OH and H2O extracts of Mentha Haplocalyx Brig., Et OH and H2O extracts of Fructus Canarii. And they reduced the proliferation and TRAP activity of osteoclasts in a dose-dependent manner. Among them, three extracts at 1 μg/m L had favorable inhibitory effects on proliferation and TRAP activity of osteoclast, including H2O extract(93%, 308%) of Mentha Haplocalyx Brig., Et OH extract(87%, 292%) and H2O extract(83%, 297%) of Fructus Canarii., which possessed the stronger anti-osteoporosis activity. Moreover, it provided scientific basis for exploring hygienical function of anti-osteoporosis of edible traditional Chinese medicine.
