臭牡丹对SNI诱导的神经病理性痛大鼠模型痛敏行为的影响 被引量:2

Effects of Clerodendrum Bungei Steud on Hyperalgesia Behavior in Rats with Neuropathic Pain Induced by Spared Nerve Injury

摘要 目的:观察臭牡丹(Clerodendrum Bungei Steud,CBS)对坐骨神经分支选择性损伤(spared nerve injury,SNI)诱导的神经病理性痛大鼠所致机械痛敏及热痛敏的影响。方法:雄性SD大鼠40只,随机分为空白对照组(Ctrl)、假手术组(Sham)、模型组(SNI)和给药组(30 g·kg^(-1)CBS组)。术后第7天开始,SNI+CBS组大鼠p.o给予30 g·kg^(-1)的CBS,以0.5 m L·100 g^(-1)为给药体积,连续灌胃28 d,Ctrl组、Sham组和SNI组大鼠均p.o给予同体积生理盐水。于术前1 d和术后第7、14、21、28和35 d分别测定机械缩足反射阈值(Paw withdrawal threshold,PWT)及热辐射刺激缩爪反应潜伏期(Paw Withdrawal Latency,PWL)。结果:与假手术组相比,模型组PWT在各时间点均明显降低(P<0.001);而PWL值变化无统计学差异(P>0.05)。与模型组相比,给予剂量30 g·kg^(-1)CBS干预后的第14、21、28和35 d,机械缩足反射阈值显著上升(P<0.05,P<0.01);热辐射刺激缩爪反应潜伏期有所增加,但差异无统计学意义(P>0.05)。结论:30 g·kg^(-1)CBS灌胃给药可显著减轻SNI所致神经病理性痛大鼠的机械缩足反射阈值,但对大鼠热辐射刺激缩爪反应潜伏期作用效果不明显。 Objective: To investigate the effect of Clerodendrum Bungei Steud( CBS) on hyperalgesia behavior in rats with a neuropathic pain induced by spared nerve injury( SNI). Methods: Forty male SD rats were randomized divided into control group( Ctrl),sham operation group( Sham),model group( SNI) and treatment group( 30 g·kg^(-1)group CBS) in the experiment. On the 7th day after operation,the rats of SNI + CBS group were given 30 g · kg^(-1)of CBS,and the rats were treated with 0. 5 m L · 100 g^(-1)for 28 days. The control group,Sham group and SNI Rats were given the same volume of saline. SNI model was established and then treated with CBS intragastric administration for 28 days.Paw withdrawal threshold( PWT) and Paw withdrawal latency( PWL) were measured by Von Frey filaments and thermal radiant instrument respectively on day 1 before operation and days 7,14,21,28 and 35 after operation. Results: Compared with the sham group,the PWT of the model group was significantly lower at all time points( P < 0. 001),but there was no significant difference in PWL values( P > 0. 05). Compared with the model group,30 g·kg^(-1)dose CBS intervention at 14,21,28 and 35 days after the operation PWT was significantly increased( P < 0. 05,P < 0. 01). PWL increased,but the difference was not statistically significant( P > 0. 05). Conclusion: 30 g·kg^(-1)CBS can significantly reduce the PWT in rats with neuropathic pain caused by SNI,but it is not obvious for PWL.
