羟基红花黄色素A对紫外线致人皮肤细胞系损伤的保护作用 被引量:8

摘要 目的评价羟基红花黄色素A(HSYA)对紫外线(UVA、UVB)辐射损伤人表皮细胞(Hacat)和皮肤成纤维细胞(ESF)的保护作用。方法建立体外紫外线辐射损伤模型,采用MTT法检测培养皮肤细胞活性,分别采用生化测定与ELISA法检测不同剂量HSYA对紫外损伤模型中活性氧(ROS)和金属基质蛋白酶-1(MMP-1)表达水平的影响。结果 UVA、UVB照射后,ESF、Hacat细胞活性显著下降,ROS、MMP-1表达水平明显上升。细胞培养液加入10,20和40μmol·L-1的HSYA,可不同程度降低UVA、UVB处理后ESF和Hacat细胞中ROS与细胞外MMP-1表达水平。结论 HSYA可抑制紫外线诱导ESF、Hacat细胞氧化损伤,对抗皮肤光老化具有一定的保护作用。 Objective To evaluate the photo-protective efficiency of hydroxysafflower yellow A(HSYA)on human epidermal cells(Hacat)and human fibroblast cells(ESF)damaged by ultraviolet A radiation(UVA)and ultraviolet B radiation(UVB).Methods ESF,Hacat cells were cultured and photo aging model was induced with radiation of UVA and UVB.After HSYA treatment,the cell proliferation was detected by MTT;Intracellular ROS levels were measured by using ROS Assay Kit;Matrix metalloproteinases-1(MMP-1)contents in the culture media were detected by ELISA.Results HSYA treatment at 10,20,and 40μmol·L-1 can promote the viability of skin cells,and reduce the UV-induced ROS and MMP-1level in photo damaged ESF and Hacat cells.Conclusion HSYA can inhibit oxidative damage in ESF and Hacat cells caused by UV radiation,which may have potential photo protection effects for skincare.
