microRNA-21、microRNA-135b和microRNA-141在结肠癌组织中的表达及相关性 被引量:4

摘要 目的探析结肠癌组织中microRNA-21、microRNA-135b和microRNA-141表达水平。方法入选该院2013年5月至2014年4月结肠癌手术切除标本40例,采自结肠癌组织及近癌切端结肠癌旁健康组织,进行实时荧光定量聚合酶链反应(RT-PCR),检测结肠癌组织及结肠癌旁健康组织中的microRNA-141、microRNA-135b、microRNA-21表达水平;依据国内外相关文献,将性别、年龄、淋巴结转移、肿瘤分期系统(TNM)、分化程度列为相关因素,评估其和结肠癌组织中microRNA-141、microRNA-135b、microRNA-21表达水平的相关性。结果结肠癌组织中microRNA-141、microRNA-135b、microRNA-21表达水平显著高于结肠癌旁健康组织,差异有统计学意义(P<0.05);结肠癌组织microRNA-141、microRNA-135b、microRNA-21表达水平与结肠癌是否发生淋巴结转移、TNM分期等指标密切相关,差异有统计学意义(P<0.05)。结论结肠癌组织中miRNA-141、miRNA-135b、miRNA-21表达水平过高,与结肠癌是否发生淋巴结转移、TNM分期密切相关。 Objective To investigate the expression level of microRNA-21,microRNA-135 band microRNA-141 in colon cancer tissue.Methods Totally 40 cases of colon cancer specimen from surgical resection in our hospital from May 2013 to April 2014 were selected,40 healthy colon tissues from people who entered our hospital at the same period for physical examination were selected.The expression level of microRNA-21,microRNA-135 band microRNA-141 were determined using real time PCR.Chosed sex,age,lymph node metastasis,TNM staging,differentiation degree as related factors by literatures published in home and abroad,assessed the correlation between the level of microRNA-141,microRNA-135 b,microRNA-21 and its expression in colorectal cancer.Results The expression level of microRNA-21,microRNA-135 band microRNA-141 in colon cancer tissues were remarkably higher than those in healthy tissues,the difference was statistically significant(P<0.05).The expression levels of microRNA-141,microRNA-135 band microRNA-21 in colon cancer tissues were closely related to the occurrence of lymph node metastasis,progression and stage of colon cancer,and the difference was statistically significant(P<0.05).Conclusion The expression of microRNA-21,microRNA-135 band microRNA-141 are upregulated in colon cancer tissue,which is highly correlated with the lymphatic metastasis and progressiveness of colon cancer.
