摘要
为了建立以草甘膦为选择标记且具有高效筛选效率的棉花遗传转化体系,研究选取WC,R15和新陆早45号这3个具有成功再生体系陆地棉的下胚轴为试验材料,分别以选择压2.5,5,10,20,50,100 mg/L的草甘膦质量浓度梯度进行草甘膦敏感性试验。结果表明,WC,R15和新陆早45号的最适抗草甘膦质量浓度分别是5,20,10 mg/L。说明不同陆地棉品种下胚轴具有不同的草甘膦抗性,选择合适浓度的草甘膦是获取阳性转化体的关键之一,研究为提高以草甘膦为选择标记的棉花转基因试验的筛选效率提供了技术支持。
For developing a transgenic cotton transformation system, in which glyphosate was used as a selective agent in high screening efficiency, the research adopted hypocotyl from three varieties of WC, R15 and Xinluzao 45 of upland cotton as experimental materials, and designed glyphosate solutions of six concentrations of 2.5, 5, 10, 20, 50, 100 mg/L to carry out glyphosate susceptibility tests. The results showed that optimum concentrations of glyphosate of WC, R15 and Xinluzao 45 resist were 5, 20, 10 mg/L, respectively.The research explained that hypocotyl of different upland cottons had differrent resistance to glyphosate. Selecting the appropriate concentration of glyphosate was one of the keys to obtain the positive transformation. So, the research provides a technical basic for cotton genetic transformation, aim at improving the screening efficiency of transgenic cotton with glyphosate as the selective marker.
