摘要
从不同角度评价氢化可的松诱导的SD大鼠肾阳虚模型,以验证此造模方法的可行性。雄性SD大鼠16只,随机分为对照组和模型组,每组8只。氢化可的松后肢肌肉注射造模15d,期间观察大鼠体征变化并测体质量、体温。15d后,心脏采血,脱颈处死大鼠,取相应脏器测质量并计算胸腺、脾脏脏器指数;ELISA方法检测大鼠血浆环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP),血清三碘甲状腺原氨酸(T3)、甲状腺素(T4)、雌二醇(E2)、睾酮(T)、促肾上腺皮质激素(ACTH)、皮质酮(CORT)、生长激素(GH)含量。结果显示:与对照组相比,模型组大鼠体质量显著减轻(P<0.01),体温显著下降(P<0.01),脏器指数显著降低(P<0.05),血浆cAMP含量显著降低(P<0.01)、cGMP含量显著升高(P<0.01),血清T3、T4、E_2、T、ACTH、CORT、GH含量显著降低(P<0.05)。结果表明:氢化可的松诱导大鼠肾阳虚模型的方法是可行的。
The insufficiency of kidney-YANG model induced by hydrocortisone in SD rats was eval- uated from different angles. A total of 16 male SD rats were randomly divided into two groups, control group and model group,8 rats of each. Hydrocortisone was injected into the hind limb for 15 d,and the body signs,body mass and body temperature were observed during the course. After that,rats were sacrificed after collecting the heart blood, the corresponding organs weight were measured, then calculate the thymus and spleen organ index; ELISA method was used for detection of plasma cAMP, cGMP, serum T3, T4, E2, T, ACTH, CORT, GH. The results showed that com- pared with the control group, body quality and temperature of rats in model group reduced signifi- cantly (P〈0. 01),viscera index decreased significantly (P〈0.05), plasma cAMP content de- creased significantly (P〈0.01), cGMP increased significantly(P〈0.01), and the content of serum T3, T4, E2, T, ACTH, CORT, GH decreased significantly (P〈0.05). The results showed that the insufficiency of kidney-YANG model induced by ihydrocortisone was feasible.
