摘要
以采自贵州凯里小高山七叶一枝花为材料,设计不同培养基与表面消毒方法进行七叶一枝花块茎内生真菌的分离,并利用高效液相色谱法对七叶一枝花内生真菌产生的薯芋皂甙元的含量进行了比较,结果共筛选出9株属于4属的七叶一枝花块茎内生真菌.然后采用正交试验的方法对不同内生真菌培养方法进行了比较,结果发现以开花期七叶一枝花块茎为材料、采用75%乙醇进行表面消毒15 min、在PDA培养基中加入4 g/L七叶一枝花粉末、培养8 d后分离得到的内生菌产薯芋皂甙元的能力最强.
The study took Paris polyphylla Smith that came from Xiaogaoshan of Kaili in Guizhou as the material to separate endophytic fungi by designing different medium and surface disinfection method. The contens of diosgenin in different endophytic fungi were compared by high performance liquid chromatography (HPLC). Nine endophytic fungi were separated from tuber of Paris polyphylla Smith, them belonged to four different genera. Different cultural method of endophytic fungi were compared through the orthogonal experimental method, the result showed that endophytic fungi may produce the most diosgenin by combination method of Paris polyphylla Smith in flowering period, using 75% ethanol sterilization for 15 min, add 4 g/L shattered tuber in PDA medium and cultivating eight days.
