摘要
为了提高蓝舌病病毒(BTV)的分离效率,2015年在师宗县设置监控点,选择蓝舌病抗体阴性的10头牛和5只羊作为监控动物,5—10月份每周采血1次,每次每头动物分别采集常规全血、EDTA抗凝血和肝素抗凝血各1份,检测蓝舌病抗体、蓝舌病病毒核酸并进行病毒分离。结果表明:蓝舌病病毒核酸阳性可以持续4~5周,蓝舌病抗体阳性可以持续5~6周。BTV核酸开始转为阳性到核酸转阳后的第1周和第2周采集的肝素抗凝血,或者蓝舌病抗体转阳前1周到蓝舌病抗体转阳和蓝舌病抗体转阳后1周采集的肝素抗凝血BTV分离效果最佳。说明通过蓝舌病抗体检测或蓝舌病病毒核酸检测确定分离蓝舌病病毒最佳分离时间是可行的。
A study was conducted to improve the efficiency of Bluetongue virus isolation. The sentinel herd for monitoring bluetongue were established in 2015. It included antibody negative 10 cattle and 5 goats. From May to October, blood samples were taken once a week. The blood samples were collected from animals mixed with heparin anticoagulant and EDTA anticoagulant for bluetongue antibody detection, Bluetongue virus nucleic acid detection and virus isolation, respectively. The results showed that, the positive BTV nucleic acid persisted a period of 4 to 5 weeks, and the positive BT antibody persisted a period of 5 to 6 weeks. The best opportunity of BTV isolation was the first week and second week after positive BTV nucleic acid, or one week before and after positive BT antibody, then the beparin anticoagulant blood samples were collected. It was concluded that the optimal separation opportunity of BTV by the BT antibody or nucleic acid detection was feasible.
