摘要
为了研究从健康7日龄仔猪盲肠中筛选出的一株益生菌菌株的体外抗传染性胃肠炎病毒(TGEV)的活性,并初步分析其抗病毒机制,试验采用形态观察、16S rRNA基因扩增,并结合MTT法观察细胞病变情况对其进行了研究。结果表明:分离鉴定的益生菌属于植物乳杆菌(暂命名为LP-1),不同接入顺序的培养上清液对TGEV增殖均有抑制作用,其中培养上清液预处理组效果优于后处理组,培养上清液与TGEV同时作用组效果最差。说明培养上清液对TGEV在体外培养猪睾丸(ST)细胞模型上的增殖能产生抑制作用,并与其不同接入顺序有关。
To study the antiviral effectof a probiotic strain which screened and isolated from the cecum of healthy piglets at 7 days old against porcine Transmissible gastroenteritis virus (TGEV)and to analyzepreliminarilyits antiviralmechanism, the methods were carried out in this study including morphological observation, 16S rRNA gene amplification, and MTT for cytopathic effectdetection to confirm the isolatedstrain. The re- sults showed that the isolated probiotie strain was identified and belonged to Lactobacillus plantarum sp. ( named LP - 1 ). LP - 1 cultured super- natants withdifferent treatmentseould inhibit TGEV proliferation. The antiviral effect of LP - 1 in pretreatment group was better than that in post treatment group andthe worsteffect was in co - incubation group of LP - 1 containedsupernatant and TGEV. The resuhsdemonstrated that LP - 1 containedculture supernatantsshowedthe ability to the inhibition of TGEV proliferationon ST cell modelin vitro, and its antiviral effect was in relation to different treatments.
