摘要 为了研究根皮素对LPS诱导的小鼠乳腺组织的炎症变化,并证明根皮素对乳腺炎具有保护作用,试验应用乳导管内灌注LPS建立小鼠乳腺炎模型,进行H.E.染色、ELISA检测、Western-blott检测和髓过氧化物酶(MPO)检测等研究根皮素对乳腺组织的保护作用。结果表明:LPS能使乳腺组织产生病理学损伤,促进白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的产生,增加组织MPO活性,增强NF-κB信号通路蛋白的表达,而根皮素能够抑制LPS诱导的小鼠乳腺组织的炎症变化。说明根皮素对LPS诱导的小鼠乳腺炎具有一定保护作用。 To study the effect of phloretin onthe inflammatory changes of LPS - induced mastitis in miceand to prove phloretin's protective effect against mastitis, micemastitis model was established in this experiment via pouring LPS into mammary duct. The protective effect of phloretinon- micemammary tissue was evaluated using several methods including H. E. staining, ELISA, Western - blottand MPO detection. The results showed that LPS could lead to pathological injury on mammary tissue in mice, and could increase the levels of cytokines IL -1β, IL -6 ,TNF - α, and also could increase MPO activity in mammary tissue and enhance the expression of NF - KB signaling pathway. Howeverphloretin could inhibitinflammatory changesof LPS - induced mastitis in mice. The results indicated thatphloretin could play a certain role in the protection against LPS -induced mastitis in mice.
