桔梗的组培离体再生 被引量:3

Tissue Culture and Regeneration of Platycodon grandiflorus in vitro

摘要 以桔梗的子叶和下胚轴为外植体,采用离体培养的方法,研究了诱导芽的频率,找出较为理想的外植体类型和相应的培养条件,以期建立高效的桔梗组培再生体系,为桔梗优良株系的繁育和利用提供有效途径。结果表明:桔梗的子叶、下胚轴均可成功诱导出再生芽,其中以子叶诱导率最高,最适合的培养基为MS+1.0 mg·L^(-1) BA+0.5 mg·L^(-1)IAA,在MS+0.5mg·L^(-1)?BA+1.0mg·L^(-1)?IAA的培养基上桔梗再生芽的生长长度最高。因此,桔梗的子叶是最理想的外植体类型,相应的分化培养基组合为MS+1.0mg·L^(-1)BA+0.5mg·L^(-1)?IAA,伸长培养基组合为MS+0.50mg·L^(-1)?BA+1.0mg·L^(-1)?IAA。 Cotyledons and hypocotyls of Platycodon grandiflorus were used as explants to analyze the bud induction frequencies of them by in vitro culture method,so as to find out the ideal plants types and the corresponding culture conditions.In order to establish an efficient regeneration system of tissue culture of Platycodon grandiflorus and provide an effective method for the breeding and utilization of fine strains.The results showed that both the cotyledons and hypocotyls of Platycodon grandiflorus regenerated buds successfully.Cotyledons′ induction rate was the highest,the most suitable medium for the MS+ 1.0 mg·L-1 BA+0.5 mg·L-1 IAA,the highest growth of Platycodon grandiflorus regeneration bud length in the medium of MS+0.5 mg·L-1BA+1.0 mg·L-1 IAA.Thus,the cotyledon explants of Platycodon grandiflorus was the most ideal explant type,the corresponding differentiation medium was MS+ 1.0 mg·L-1 BA+0.5 mg·L-1 IAA,elongation medium combination MS+0.50 mg·L-1 BA+1.0 mg·L-1 IAA.
